新风胶囊对类风湿关节炎患者CD4^＋CD25^＋CD127^lo调节性T细胞变化的影响

目的：探讨新风胶囊对类风湿关节炎（Rheumatoid arthritis，RA）患者外周血CCD4^＋CD25^＋CD127^lo调节性T细胞（CD4^＋CD25^＋CD127^loTreg）变化的影响。方法：40例RA活动期患者随机分为正清风痛宁胶囊组（对照组）和新风胶囊组（XFC组），每组20例，同时设正常组20例。XFC组和对照组均治疗1个疗程，采用流式细胞术检测RA活动期患者治疗前后以及正常组外周血中CD4^＋CD25^＋CD127^loTreg的数量及其在CD4^＋T淋巴细胞中所占的比例，同时观察临床症状．DAS28评分及部分实验室指标（ESR、CRP、RF）的变化。结果：RA活动期患者外周血CD4^＋CD25^＋CD127^loTreg数量及其占CD4^＋T淋巴细胞的百分比均低于正常组；XFC组在总有效率、改善RA关节症状及部分实验室指标方面与对照组作用相似，但在上调CD4^＋CD25^＋CD127^loTreg表达水平、降低DAS28分值方面显著优于对照组。结论：XFC治疗类风湿性关节炎作用机制可能与上调CD4^＋CD25^＋CD127^loTreg表达水平，促进免疫调节、维持免疫耐受有关。

Effect of Xinfeng Capsule on the Content of Peripheral Blood CD_4~+CD_(25)~+CD_(127)~(lo) Regulatory T Cell of Rheumatoid Arthritis Patients in Active Stage

Objective:To observe the effect of Xinfeng capsule on the content of peripheral blood CD4+CD25+CD127lo regula-tory T cell(Treg)of rheumatoid arthritis(RA)patients in active stage. Methods:40 RA patients in active stage were ran-domly divided into Xinfeng capsule group (XFC group)and Zhengqing Fengtongning capsule group (control group),20 healthy persons were collected as normal group.Before and after treatment, articular symptoms were observed and DAS28 value was calculated in RA patients;ESR、CRP and RF were measured,the content of peripheral blood CD4+CD25+CD127lo Treg and its percentage in CD4+ T cell were detected by flow cytometry in all groups. Results:The content of CD4+CD25<'+> CD127lo Treg and its percentage in CD4+ T cell were lower in RA patients than those in healthy person(P<0.01);the content of CD4+CD25+CD127lo Treg was higher and DAS28 value was lower in XFC group than those in control group(P<0.01). Con-clusion:XFC could promote immunoloregulation and maintain immune tolerance through upregulating of CD4+CD25+CD127lo expression levels.
