脊髓损伤后碱性成纤维细胞生长因子的表达及意义

摘    要:目的 观察脊髓损伤 (SCI)后大鼠后肢功能恢复情况 ,以及损伤后不同时间碱性成纤维细胞生长因子(b FGF)表达的变化及意义。 方法 取雌性 SD大鼠 4 0只随机分为 5组 (n=8) :正常对照组、伤后 2 ,5 ,1 2和 1 8d组。 Allen法致伤脊髓。免疫组织化学和图像分析方法观察神经细胞 b FGF表达变化。大鼠综合行为评分法(CBS)对各级神经功能评分。 结果  (1 )在行为观察中 ,发现大鼠脊髓损伤后有自行恢复倾向 ,损伤后 1 8d后肢功能恢复 6 8%。 (2 ) b FGF表达 :2 ,5 ,1 2 d b FGF表达量及 b FGF阳性细胞数目呈进行性增加 ,但在 1 8d开始下降。损伤后的前 1 2 d,各组 b FGF灰度值与 CBS评分有显著的相关性 (r=- 0 .95 98,P<0 .0 1 )。 结论  (1 )急性脊髓损伤后脊髓有自我修复的倾向 ;(2 ) b FGF在急性脊髓损伤后早期可能对脊髓的再生和自我修复起重要作用

Abstract:Objective To observe the recovery of hindlimb function and the expression of basic {fibroblast} growth factor(bFGF) after spinal cord injury(SCI) and its significance. Methods 40 female SD rats were randomity divided into the five groups: the groups of 2, 5, 12, 18 day after spinal cord injury and control group(n=8). Spinal cord was injured with Allen's weight|dropping model. Immunohistochemical technique and image analysis technique were used to detect the expression of bFGF in spinal cord tissue, and the neurological function of spinal cord was evaluated according Gale combined behavioral score(CBS). Results (1)In behavior examination, there was about 68% self|recovery of hindlimb function in the injured rats. (2)The expression of bFGF and the number of bFGF positive cell increased from 2~12th day, and decreased at 18th day. bFGF was significantly negatively correlated with CBS(r={-0 9598},P<0 01). Conclusion (1)There was a tendency of self|repairing after spinal cord injury. (2)bFGF may play an important role in self|repairing and regeneration in spinal cord injury.
