异氟醚对新生大鼠认知功能和突触素表达的影响

摘    要:目的观察异氟醚对新生大鼠认知功能的影响及其机制和海马突触素(synaptophysin)的影响。方法新生1周Wistar大鼠40只,雌雄不拘,随机分为5组(n=8):对照组(A组)吸入空气;B、C组分别吸入1.8%和1.2%异氟醚6 h,D、E组分别吸入1.8%、1.2%异氟醚3次,1次/d,2 h/次。吸入异氟醚后3周进行认知功能测试,包括旷场实验和Morris水迷宫测试,然后取大鼠海马组织行免疫组织化学染色,测定突触素表达。结果吸入异氟醚可导致大鼠认知功能降低,海马突触素表达增加(P〈0.05),吸入高浓度异氟醚后海马突触素表达增加更明显,单次吸入和多次吸入异氟醚海马突触素表达无影响(P〉0.05)。结论异氟醚可降低新生大鼠认知功能,并上调海马突触素表达。

Abstract:Objective To investigate the effects and mechanism of isoflurane on cognitive function and synaptophysin at hippocampus in neonatal rats.Methods Forty neonatal 1 week Wistar rats were randomly divided into five groups(n=8): Group A breath normal air as control;Group B and Group C were anesthetized constantly with 1.8% and 1.2% isoflurane for 6h respectively;Group D and Group E were anesthetized with 1.8% and 1.2% isoflurane for 2h and repeated three times within 3 days respectively.Open field and Morris water maze were tested after three weeks.Meanwhile,Immunohistochemistry technique was implicated to observe the changes of synaptophysin at hippocampus.Results Isoflurance decreased cognitive function assessed by Morris water maze(P<0.05) and increased the expressions of synaptophysin at hippocampus compared with that of control group(P<0.05),especially in 1.8% isoflurane anesthetized group(P<0.05),but there was no significant difference between single and repeated anesthetic style(P>0.05).Conclusion Isoflurance can decrease cognitive function and this effects maybe induced by the increase of the expressions of synaptophysin at hippocampus in neonatal rats.
