空间辨别性学习记忆活动对大鼠额叶SDF-1免疫阳性细胞的影响

摘    要:
目的讨大鼠空间辨别性学习记忆活动对额叶基质细胞衍生因子-1(SDF-1)免疫阳性细胞的影响。方法用水迷宫训练SD大鼠建立空间辨别性学习记忆模型,用免疫组化方法和图像分析技术检测大鼠额叶SDF-1免疫阳性细胞的形态变化。结果对照组大鼠额叶可见少量圆形或椭圆形SDF-1免疫阳性细胞分布在外颗粒层、外锥体层、内颗粒层和内锥体细胞层,分子层和和多形层少见阳性细胞。阳性细胞的胞浆淡黄色,颗粒状免疫反应产物附于细胞膜内面。游水组大鼠额叶皮质SDF-1免疫阳性细胞不论分布还是形态均未见与对照组的有明显差异;模型组训练7d的大鼠额叶SDF-1免疫阳性细胞的形态和分布与对照组的比未见明显差异。训练14d和21d的再细胞形态上以锥体形或三角形为主,在分布上,额叶II区较I区的密集,在SDF-1表达上训练21d的比训练7d和14d的明显增多。结论大鼠额叶内SDF-1免疫阳性细胞可能参与空间辨别性学习记忆活动引致的形态学可塑。

Abstract:
Objective To explore effects of the activity of spatial learning and memory on cell morphology of SDF-1 immuno-positived and SDF-1 expression in frontal lobe cortex in rats. Methods spatial learning and memory activities of SD male rats were trained with water maze. Immunohistochemistry and imaging analyzing system were used to study the location , shape and number changes of SDF-1-positive cells in rat frontal lobe cortex. Results The positive cells of SDF-1immunoreactive in frontal lobe cortex distributed in extemal granular cell layer, extemal pyramidal layer, intemal granular layer and intemal pyramidal layer, but less in the molecular layer and polymorphic layer. The positive cell body showed round and oval in shape and endochylema showed buffy in control group, granular immune reaction product adhesion to the inner cell membrane surface ; The cell characters in swimming group was as same as those in control group ; there is no difference about the cell morphology and distribution between control group and model rats trained 7 days. In the model trained 14 days and 21 days , the shape of the positive cell was mainly pyramid and triangele, the immunoreactivity in the areas was intensive than the Ⅰ areas. The expression of SDF-1 was much more in trained 14 days and 21 days rats than trained 7 days and control group. Conclusion SDF-1 maybe correlates with the spatial learning and memory activity and maybe take part in the cellular morphological plasticity in frontal lobe cortex.
