大豆皂甙AI对心肌作用的单钙通道分析及ESR谱研究

摘    要:利用细胞贴附式膜片钳技术,在Wistar大鼠单个心室肌细胞上记录了T、L、B型钙通道的单通道活动,大鼠皂甙单体AI20μmol/L显著抑制三型钙通道的活动,使其开放概率减少,开放时间缩短,对通过通道的离子流幅值无明显影响,用电子自旋共振法测定了培养心肌细胞的自由基含量。大豆皂甙单体AI2μg/ml显著抵消黄嘌呤0.42mmol/L-黄嘌呤氧化酶5.3nmol/L所致的自由基含量增多。

Abstract:Single channel activity of type T,L,B Ca 2+ channels were recorded on single ventricular myocardiocytes of Wistar rats,using cell attached configuration of patch clamp technique.Soyasaponin monomer AI 20 μmol/L inhibited the activities of the three types of Ca 2+ channel significantly.The open state propability was decreased and the open time was shorened.2 μg/ml significantly antagonized the increase of the free radical content induced by xanthine 0.42 mmol/L and Xanthic oxidase 5.3 nmo/L
