壮元扶骨逍遥汤对家兔膝骨关节炎血清SOD和关节液IL-1含量的影响

摘    要:目的 观察壮元扶骨逍遥汤对家兔膝骨关节炎模型血清超氧化物歧化酶(SOD)和关节液中白细胞介素-1(IL-1)含量的影响,并探讨其作用机制.方法 将新西兰白兔36只,雌雄不分,随机分成4组,即正常组(N组)、模型对照组(M组)、壮元扶骨逍遥汤组(F组)、布洛芬组(P组),各8只,于相同温度、湿度下分别单笼喂养,在治疗前、中、后分别提取家兔的血清和关节液,采用免疫组织化学法检测血清SOD和关节液IL-1的含量.结果 治疗前比较,M、F、P组SOD水平明显降低(P<0.05),IL-1水平明显升高(P<0.05);治疗中、后期比较,F组SOD水平明显升高(P<0.05),P组无明显变化(P>0.05),F、P组IL-1水平明显降低(P<0.05).结论 壮元扶骨逍遥汤可提高血清SOD水平,保护关节软骨,提高软骨细胞功能,促进软骨修复;降低关节液IL-1水平,减轻炎症对关节软骨的破坏及所产生的疼痛.

Experimental investigation on treating osteoarthritis of the knee with Zhuangyuanfuguxiaoyao decoction

Abstract:Objective To observe the influence of Zhuangyuanfuguxiaoyao decoction on SOD in blood serum and IL-1 in synovial fluid of knee osteoarthritis rabbit model, and reveal the mechanism of action. Methods 36 well-being New Zealand white rabbits were randomly divided into 4 group, they are normal group(group N), model reference group(group M), Zhuangyuanfuguxiaoyao decoction group(group Z), ibuprofen group(group P), 8 rabbits in each group. The rabbits blood serum and synovial fluid were extracted in pre-treatment, meta- treatment and post- treatment, and SOD and IL-1 levels were detect with the immunohistochemistry. Result The concentration of SOD in group M, group Z and group P before treatment could markedly reduce compared with that in group N (P<0.05), and the concentration of IL-1 increased (P<0.05). The concentration of SOD meta- treatment and post- treatment group Z was higher than those pre-treatment (P<0.05), but group P there was no significant difference (P>0.05). The concentration of IL-1 meta- treatment and post- treatment group Z and group P were lower than those pre-treatment (P>0.05). Conclusion Zhuangyuanfuguxiaoyao decoction can enhance the level of blood serum SOD by activating activity of SOD, and prevent damage of arthrodial cartilage from oxygen radical by catalyzing dismutation reaction of oxygen radical. Zhuangyuanfuguxiaoyao decoction can reduce the level of IL-1 in synovial fluid and effectively by inhibition of macrophage secretion.
