糖周通对糖尿病周围神经病变大鼠周围神经功能和血清炎症标记物的影响

摘    要:目的探讨糖周通对链脲佐菌素(STZ)诱导的糖尿病周围神经病变大鼠周围神经功能、糖代谢、血尿酸及炎症因子的影响。方法取SPF级雄性SD大鼠80只,随机抽取20只作为正常组,常规喂养;其余大鼠采用高脂饲料和腹腔注射链脲佐菌素方法诱导2型糖尿病周围神经病变大鼠模型。将成模大鼠随机分为模型组、弥可保组、糖周通组,每组20只。弥可保组大鼠灌胃弥可保混悬液175μg/(kg·d),糖周通组大鼠灌胃糖周通煎剂25 g/kg,正常组和模型组大鼠灌胃相同容量的生理盐水,均1次/d,连续8周。末次灌胃结束后24 h,检测各组大鼠坐骨神经传导速度、空腹血糖、空腹胰岛素、胰岛素敏感指数、血尿酸、血清白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平。结果与正常组比较,模型组大鼠坐骨神经的运动神经传导速度和感觉神经传导速度均明显减慢(P均<0.05),空腹胰岛素水平、胰岛素敏感指数均明显降低(P均<0.05),空腹血糖、血尿酸、IL-6及TNF-α水平均明显升高(P均<0.05)。弥可保组、糖周通组大鼠坐骨神经的运动神经传导速度和感觉神经传导速度均明显快于模型组(P均<0.05),且糖周通组均明显快于弥可保组(P均<0.05);糖周通组大鼠空腹血糖、血尿酸水平均明显低于模型组和弥可保组(P均<0.05),空腹胰岛素、胰岛素敏感指数均明显高于模型组和弥可保组(P均<0.05);弥可保组、糖周通组大鼠血清IL-6、TNF-α水平均明显低于模型组(P均<0.05),且糖周通组均明显低于弥可保组(P均<0.05)。结论糖周通能有效改善糖尿病周围神经病变大鼠的周围神经功能和糖代谢障碍,降低血尿酸水平,机制可能与降低血清炎症因子水平相关。

Abstract:Objective It is to investigate the effects of Tangzhoutong on peripheral nerve function,glucose metabolism,blood uric acid and inflammatory factors in rats with streptozotocin(STZ)-induced diabetic peripheral neuropathy.Methods Eighty SPF male SD rats were selected in which 20 ones were randomly selected as the normal group conventionally fed.The remaining rats were induced by high-fat diet and intraperitoneal injection of streptozotocin to establish type 2 diabetic peripheral neuropathy.The model rats were randomly divided into a model group,a micobala group,and a Tangtangtong group,with 20 rats in each group.The rats in the micobo group were given 175μg/(kg·d)of micobo suspension by gavage,the rats in the Tangzhoutong group were given tangtangtong decoction 25 g/kg by gavage,the rats in the normal group and model group were given the same volume of normal saline,all once/d for 8 weeks.In 24 hours after the last gavage,the sciatic nerve conduction velocity,fasting blood glucose,fasting insulin,insulin sensitivity index,blood uric acid,serum interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were measured in the rats of each group.Results Compared with the normal group,the motor nerve conduction velocity and sensory nerve conduction velocity of the sciatic nerve in the model group were significantly reduced(both P<0.05),and the fasting insulin level and insulin sensitivity index were significantly reduced(all P<0.05),fasting blood glucose,serum uric acid,IL-6 and TNF-αlevels were significantly increased(all P<0.05).The motor nerve conduction velocity and sensory nerve conduction velocity of the sciatic nerve in the micobo group and Tangzhoutong group were significantly faster than those in the model group(P<0.05),and the Tangzhoutong group was significantly faster than the micobao group(P<0.05);the fasting blood glucose and serum uric acid levels of the rats in the Tangzhoutong group were significantly lower than those of the model group and micobo group(both P<0.05),and the fasting insulin and insulin sensitivity indexes were significantly higher than those of the model group and micobo group(All P<0.05);the levels of serum IL-6 and TNF-αin the rats of micobo and Tangzhoutong group were significantly lower than those of the model group(P<0.05),and the levels of Tangzhoutong group were significantly lower than those of micobo group(P<0.05).Conclusion Tangzhoutong can effectively improve peripheral nerve function and glucose metabolism disorders,reduce blood uric acid levels in diabetic peripheral neuropathy rats,and the mechanism may be related to lowering serum inflammatory factor levels.
