固本消渴汤对2型糖尿病大鼠骨骼肌GLUT-4和ABCA1mRNA表达的影响

目的：观察固本消渴汤对2型糖尿病大鼠骨骼肌葡萄糖转运蛋白-4（GLUT-4）及脂肪组织三磷酸腺苷结合盒转运体A1（ABCA1）表达的影响。方法：雄性Wistar大鼠随机分为正常对照组和造模组。造模组成功后再分为模型对照组、阳性药对照组和中药治疗组，分别给予生理盐水和固本消渴汤药液灌胃。定期定时监测空腹血糖、OGTT、FBG、FINS。第12周末，取后肢骨骼肌，检测GLUT-4和ABCA1的基因与蛋白的表达。结果：固本消渴汤组能降低BG、TC、TG的水平，增强糖耐量能力，明显增加糖尿病大鼠骨骼肌 GLUT-4和 ABCA1基因与蛋白的表达。结论：固本消渴汤能调节糖尿病大鼠糖脂代谢，增加糖尿病大鼠骨骼肌组织GLUT-4和ABCA1的表达。

Effects of Guben Xiaoke decoction on GLUT-4 and ABCA1 mRNA expression in type 2 diabetic rats

Objective： To investigate the effects of Guben Xiaoke decoction on the GLUT-4 and ABCA1 expression in skeletal muscle of type 2 diabetic rats. Methods： Male Wister rats were randomly divided into the normal group and the model group. Then the model group divided into model control group, positive medicine control group and TCM group after the model success, they were given normal saline and Guben Xiaoke decoction by lavage. Fasting blood-glucose, OGTT, FBG, FINS were regularly monitored, gene and protein expression of GLUT-4 and ABCA1 in the hindlimb skeletal muscle were tested at the end of the twelfth week. Results： The Guben Xiaoke decoction group can reduce the BG, TC, TG level, enhance the capacity of glucose tolerance, and markedly increase effects of the GLUT-4 and ABCA1 in skeletal muscle of rats with diabetes gene and protein. Conclusion： Guben Xiaoke decoction can prevent diabetes vascular complicationgs by regulation of glycolipid metabolism, increase the expression of GLUT-4 and ABCA1.
