摘    要:
目的 研究淫羊藿总黄酮(Total flavonoid extract of Eimedium sagittatum,TFE)对大鼠骨髓间充质干细胞(Mysenchymal stem cells,MSCs)增殖和成骨性分化的影响。方法将TFE和含淫羊藿总黄酮大鼠血清(Serum of rats administered TFE,SES)分别以不同浓度加入大鼠MSCs培养液中,进行细胞增殖和成骨性分化的分析。细胞增殖分析采用MTT法,成骨性分化则检测碱性磷酸酶、骨钙素、钙盐沉积量等分化指标。结果TFE直接加入培养液对:MSCs增殖无明显影响,SES则强烈刺激细胞增殖,成倍增加碱性磷酸酶染色阳性的克隆数。TFE不影响MSCs的成骨性分化,但SES显著提高碱性磷酸酶活性、骨钙素分泌量和钙盐沉积量。结论 淫羊藿总黄酮活性代谢产物强烈刺激MSCs的增殖和成骨性分化,是淫羊藿总黄酮抗骨质疏松症的重要机制。

Abstract:
Objective To investigate the effects of total flavonoid extract of Epimedium sagittatum (TFE) on the proliferation and differentiation of rat marrow mesenchymal stem cells (MSCs) . Methods TFE and the serum of rats administered TFE (SES) were added into the culture medium of newborn rat MSC, Their effects on cell proliferation and osteogenic differentiation were studied by MTT and the analysis of differentiation indices. Results TFE had no appreciable effect on cell proliferation and differentiation at any concentration, while SES strongly stimulated cell proliferation, increased the number of colonies stained positive for alkaline phosphatase. SES also significantly improved osteogenic differentiation indices including alkaline phosphatase activity, osteocalcin secretion and calcium deposition level. Conclusions It is the metabolites of TFE but not the extract itself that enhance bone formation by increasing the number of MSCs and inducing their differenation into osteoblasts. It partly explains the anti-osteoporosis mechanism of TFE.
