裙带菜多糖对大鼠黑质内多巴胺能神经元6-羟基多巴胺致损伤的保护作用

摘    要:目的探讨裙带菜多糖(UPPS)对6-羟基多巴胺(6-OHDA)制备的大鼠帕金森(PD)模型动物的多巴胺(DA)能神经元的保护作用。方法将36只Wistar大鼠随机分为对照组、UPPS+6-OHDA组、6-OHDA组。对照组连续生理盐水灌胃30d;UPPS+6-OHDA组,裙带菜多糖(400mg/kg)灌胃3d后,内侧前脑束(MFB)注射6-OHDA(3.6mg/ml),随后连续裙带菜多糖(400mg/kg)灌胃26d;6-OHDA组,以生理盐水代替裙带菜多糖,其他处理同UPPS+6-OHDA组。检测各组大鼠纹状体(Str)内多巴胺、3,4-二羟苯乙酸(DOPAC)、高香草酸(HVA)的含量,6-OHDA损毁侧大鼠黑质(SN)酪氨酸氢化酶(TH)阳性细胞数和铁染色阳性细胞数,大鼠黑质SOD/β-actin灰度值。结果 UPPS+6-OHDA组DA、DOPAC的含量低于对照组(P

THE PROTECTIVE EFFECTS OF UNDARIA PINNATIFIDA POLYSACCHARIDES ON 6-HYDROXY DOPAMINE INDUCED DAMAGE OF SUBSTANTIA NIGRA DOPAMINERGIC NEURONS IN RATS
Abstract:Objective To investigate the protective effects of Undaria pinnatifida polysaccharides(UPPS) on dopaminergic neurons of female rats after 6-hydroxy dopamine(6-OHDA) lesions.Methods Thirty-six female Wistar rats were randomly divided into three groups: control group,UPPS+6-OHDA group,6-OHDA group.The control group was daily administered with normal saline(NS) i.g..The UPPS+6-OHDA group was administered with UPPS i.g.3 d at first,then damaged medial forebrain bundle(MFB) with 6-OHDA,and daily administered with UPPS i.g.for 26 d.UPPS were replaced by NS in 6-OHDA group.The contents of dopamine(DA),3,4-dihydroxyphenylacetic acid(DAPAC),homovanillic acid(HVA) in striatum,the numbers of tyrosine hydroxylase(TH)positive neurons,iron-staining cells,the ratio of SOD/β-actin protein levels in the substantia nigra(SN) were determined.Results The contents of DA,DOPAC in UPPS+6-OHDA group decreased compared with those in control group(P<0.05) and increased compared with that of 6-OHDA group(P<0.05).UPPS significantly inhibited 6-OHDA-induced reduction of TH positive neurons and increase of iron-staining cells(P<0.01).UPPS also significantly inhibited 6-OHDA-induced decreace in the ratio of SOD/β-actin protein levels(P<0.05).Conclusion UPPS exerts neuroprotective effect on dopaminergic cells in 6-OHDA-induced rats Parkinson model.ACTA NUTRIMENTA SINICA,2013,35(2):186-190]
