姜黄素防治慢性心力衰竭大鼠左心室胶原网络重构的实验研究

摘    要:目的 观察姜黄素对慢性心力衰竭(CHF)大鼠左室胶原网络重构的影响。方法 采用缩窄大鼠腹主动脉的方法,将大鼠随机分成假手术组、手术对照组、卡托普利组和姜黄素组,姜黄素组又分为高、中、低3个剂量组,造模后每周测定鼠尾动脉血压,分别于造模后第12、20周观察左室质量指数、心脏血流动力学变化,采用VG染色方法检测心肌胶原含量。结果 12周末手术对照组LVEDP、LVMI、CVF-V、CVF—NV均明显强于其余各组,20周末与手术对照组相比,姜黄素各剂量组和卡托普利组大鼠LVSP均增加,LVEDP、LVMI以及CVF-V、CVF-NV的表达显著降低,姜黄素高剂量组尤为明显;不同时间段,假手术组LVEDP、LVMI、CVF-V、CVF-NV的前后表达相近,而手术对照组、卡托普利组和姜黄素各剂量组前后差别显著。结论 姜黄素具有抑制胶原网络重构的作用,可在一定程度改善心功能,延缓CHF的进程。

Abstract:Objective:The study was conducted to investigate the effects of curcumin on myocardial collagen remodeling in chronic heart failure rats.Methods:Pressure overloaded rats were established by partial abdominal aorta ligation.The rats were divided into the sham-operation group,the vehicle group,the captopril group and the curcumin group.The curcumin group was divided into the high,middle and low dose group.After operation,we detected rats' atreria caudilis blood pressure weekly and observed left ventricular mass index,cardiac hemodynamics at he 12th and 20th week.Using the VG staining detected the content of cardiac muscle collagen.Results:At the end of the 12th week,the LVEDP,LVMI,CVF-V and CVF-NV in the vehicle group were significantly increased compared with that in others groups.Compared with the vehicle group,left ventricular function in the curcumin groups and captopril group were significantly ameliorated,as indicated by decreased LVEDP,LVMI,CVF-V and CVF-NV,as if the effect of curcumin high dose group was beter.During the different periods of CHF,the LVEDP,LVMI,CVF-V and CVF-NV were no significant difference in the sham-operation,but there were significant difference in others groups.Conclusion:Curcumin can prevented collagen net remodeling and improved the cardiac function.
