血浆Gas6水平及其基因多态性与心脑血栓性疾病的相关性研究

摘    要:
目的分析血浆Gas 6水平及其基因多态性与心、脑血栓性疾病(脑梗死、脑出血、急性心肌梗死)的相关性,并探讨其可能的作用机制。方法用ELISA方法测定脑梗死组、脑出血组、急性心肌梗死组及健康对照组血浆Gas6水平,并行基因测序分析Gas 6内含子c.834+7G〉A位点的多态性频率分布。结果脑梗死组、脑出血组及急性心肌梗死组Gas 6水平显著高于对照组(P〈0.01),其Gas 6内含子c.834+7G〉A中AA基因型的Gas 6水平明显低于对照组(P〈0.01),而GA基因型组Gas 6水平明显高于对照组(P〈0.01);Gas 6基因c.834+7G〉A位点中AA基因型和等位基因A的频率显著低于对照组,与脑梗死、脑出血及急性心肌梗死呈负相关(P〈0.01)。结论血浆Gas 6水平增高与脑梗死、脑出血及急性心肌梗死相关;其c.834+7G〉A位点AA基因型和A等位基因的频率与脑梗死、脑出血及急性心肌梗死呈负相关,可能起到保护脑梗死、脑出血和急性心肌梗死的作用。

Abstract:
Objective To study the relationships of plasma Gas 6 levels and its gene polymorphism in patients with cardiac-cerebral thrombosis disease(cerebral infarction,cerebral hemorrhage and acute myocardial infarction),and to explore its possible mechanism.Method Plasma concentrations of Gas 6 was measured by enzyme-linked immunosorbent assay method and the role of Gas 6 in intron c.834 +7 G> A polymorphism frequency distribution was investigated in healthy controls and patients with cerebral infarction,cerebral hemorrhage and acute myocardial infarction.Results TThe concentration of Gas 6 was significantly higher in those patients with cerebral infarction,cerebral hemorrhage and acute myocardial infarction than in healthy controls,and the Gas 6 level in AA genotype of the Gas 6 in intron c.834 +7 G> A was significantly decreased compared with normal controls,but GA genotype is higher.The AA genotype and A allele frequency of Gas 6 c.834 +7 G> A was significantly less in patients with cerebral infarction,cerebral hemorrhage and acute myocardial infarction than in healthy controls,and was negatively correlated with cerebral infarction,cerebral hemorrhage and acute myocardial infarction.Conclusion The elevated Gas 6 plasma levels were correlated with cerebral infarction,cerebral hemorrhage and acute myocardial infarction.The AA genotype and A allele frequency were negative correlated with cerebral infarction,cerebral hemorrhage and acute myocardial infarction which may have a protective role against them.
