苦参碱对肺腺癌细胞凋亡及bcl-2、bax表达的影响

摘    要:
目的探讨中药提取物苦参碱对人肺腺癌细胞系SPC-A-1的凋亡诱导作用及其发生的机制。方法用不同浓度的苦参碱对体外培养的SPC-A-1细胞进行干预。分别采用MTT、免疫细胞化学染色、流式细胞仪检测的方法,观察其对SPC-A-1细胞的增殖抑制和凋亡诱导作用;并对凋亡相关bcl-2与bax蛋白表达的变化进行定性检测。结果经不同浓度的苦参碱处理后的SPC-A-1细胞,其生长受到明显的抑制,细胞凋亡指数随药物浓度增加而明显增加;实验组bax表达明显高于对照组。结论一定浓度的苦参碱能抑制SPC-A-1细胞的增殖,促进其凋亡;其机制可能与调控bcl-2、bax表达有关。

Abstract:
Objective It is to discuss the induction effect of Matrine which is extract of traditional Chinese drug on SPC-A-1 apoptosis in human pulmonary adenocarcinoma cell line and explore the mechanism of action.Methods SPC-A-1 cells cultured in vitro were interfered with different concentration of Matrine.The proliferation inhibition and apoptosis induction effects on SPC-A-1 cells were observed with the methods of MTT,Immunocytochemical stain and flow cytometer detected.The expression changes of bcl-2 and bax proteins correlated with apoptosis were done qualitative detection.Results The growths of SPC-A-1 cells which were processed with different concentration of Matrine were obviously inhibited.The index of apoptosis obviously increased along with increased concentration of Matrine.The expression of bcl-2 in experimental group was obviously higher than that in control group.Conclusion Determinate concentration of Matrine can inhibit proliferation and promote apoptosis of SPC-A-1 cells,and the mechanism may be correlated with regulating and controlling the expression of bcl-2 and bax.
