脂肪酸-辅酶A连接酶4的表达与胃癌细胞增殖、凋亡的关系

摘    要:目的:研究脂肪酸-辅酶A连接酶4(FACL4)在胃癌中的表达,及其与8-溴-环腺苷酸(8-Br-cAMP)调控胃癌细胞增殖、凋亡的关系。方法:采用逆转录聚合酶链反应技术,检测23例胃腺癌组织和癌旁正常胃黏膜、以及8-Br-cAMP作用SGC7901细胞后FACL4 mRNA的表达水平。应用流式细胞仪及透射电镜技术,检测2×10-5mol/L浓度的8-Br-cAMP作用48 h后SGC7901细胞的凋亡情况。结果:与正常胃黏膜相比,胃腺癌组织及SGC7901细胞中FACL4 mRNA表达水平显著上调(P<0.05),胃腺癌组织FACL4 mRNA阳性表达率为87%。2×10-5mol/L的8-Br-cAMP可抑制SGC7901细胞中FACL4的表达,从而抑制胃癌细胞增殖,诱导细胞凋亡。结论:FACL4在胃癌组织中表达增高,8-Br-cAMP作为选择性FACL4抑制剂,可抑制胃癌细胞增殖和FACL4表达,诱导胃癌细胞凋亡,可能成为胃癌诊治的一个新靶点。

Abstract:Objective:To probe the expression of fatty acid-CoA ligase 4(FACL4) in gastric adenocarcinoma,normal gastric mucosa and in SGC7901 cell lines,and to evaluate the relationship between FACL4 expression and the proliferation/apoptosis in gastric carcinoma.Methods:RT-PCR was used to analyze FACL4 mRNA expression in 23 paired tumors and normal gastric mucosa in frozen samples,and SGC7901 cell line treated with 2×10-5 mol/L 8-Br-cAMP for 48 hours.The cell apoptosis was measured by flow cytometry and transmission electron microscope.Results:FACL4 expression was remarkably up-regulated in gastric carcinomas compared with that in normal tissues(P<0.05),and the expression rate was 87% in cancerous tissues.8-Br-cAMP at 2×10-5mol/L could suppress FACL4 expression in SGC7901 cell lines,arrest the growth of grastric cancer cells and induce their apoptosis.Conclusion:The expression of FACL4 was up-regulated in gastric carcinoma,and it played an important role in the progress of gastric carcinoma.Selective inhibitor of FACL4 may provide a novel approach for diagnosis and treatment of gastric carcinoma.
