网络药理学视角下泻心汤对胃溃疡的作用机制

摘    要:目的:通过网络药理学方法,以“药物-成分-靶点-通路-疾病”网络使泻心汤治疗胃溃疡的作用机制得以具体化、精准化、可视化展示,为进一步探讨泻心汤治疗胃溃疡作用机理奠定基础。方法:在TCMSP数据库中检索泻心汤的有效成分和作用靶点,在Uniprot数据库中检索药物靶点所对应的人类基因;在GeneCards数据库中检索胃溃疡的疾病靶点;使用韦恩图获取有效成分与疾病的共有靶点;在String数据库中进行蛋白相互作用(PPI)数据分析,在Cytoscape 3.7.1中进行PPI可视化分析;使用Metascape数据库实现GO功能、KEGG通路富集分析,使用在线网络作图工具绘制高级气泡图。在Cytoscape 3.7.1中制作“药物-有效成分-关键靶点-通路”网络图。结果:泻心汤有63个有效成分、228个作用靶点,与胃溃疡有109个交集靶点,获得1 803个GO条目、176条KEGG富集通路。结论:泻心汤通过槲皮素、汉黄芩素、黄芩素、刺槐素、芦荟大黄素等有效成分调控AKT1、TP53、IL6、TNF、VEGFA等核心靶点,以及癌症通路、PI3K-Akt信号通路、MAPK信号通路、TNF信号通路等主要通路,发挥抗炎、抑酸、抗幽门螺杆菌、保护胃粘膜、防癌抑癌等作用,对胃溃疡起到治疗目的。

Abstract:Objective To specify and visualize precisely the mechanism of action of Xiexin Decoction in the treatment of gastric ulcer through the network pharmacology with the "drug-ingredient-target-pathway-disease" network, and thus to lay foundation for further exploration. Methods The active ingredients and action targets of Xiexin Decoction were searched in TCMSP database, and the corresponding human genes were searched in Uniprot database; the disease targets of gastric ulcer were obtained in GeneCards database; the common targets for drugs and diseases were obtained using Wayne diagram; Protein-Protein Interaction networks (PPI) analysis was performed in String database and PPI visualization analysis was finished using Cytoscape 3.7.1. GO function and KEGG pathway enrichment analyses were performed by Metascape database, and advanced bubble map was drawn by online drawing platform. The network diagram of "drug-active ingredient-key target-pathway" was made in Cytoscape3.7.1. Results There were 63 active ingredients in Xiexin Decoction, 228 action targets, 109 intersection targets with gastric ulcers and 1,803 GO entries and 176 KEGG enrichment pathway were obtained. Conclusion Xiexin Decoction regulates AKT1, TP53, IL6, TNF, VEGFA and other core targets as well as cancer pathways, PI3K-Akt signaling pathway, MAPK signaling pathway, TNF signaling pathway through effective components such as quercetin, wogonin, baicalein, Robinia pseudoacacia, and aloe-emodin, and plays the role of anti-inflammatory, anti-acid, anti-HP, protecting gastric mucosa, anti-cancer and anti-cancer in treating gastric ulcer.
