HI-6对梭曼染毒大鼠膈肌胆碱酯酶的重活化作用

摘    要:
目的研究HI-6对膈肌局部梭曼染毒大鼠膈肌胆碱酯酶的重活化作用。方法将大鼠麻醉后固定,沿腹正中线切开腹腔,暴露膈肌。梭曼局部染毒膈肌1和10min后,局部或全身给予HI-6,于给药后24和72h处死大鼠,取出中毒部位膈肌。用硫胆碱酶法显示终板的AChE活性,观察HI-6对胆碱酯酶的重活化作用。结果膈肌局部梭曼中毒1和10min后.局部给予HI-624和72h对局部中毒膈肌胆碱酯酶均没有重活化作用.在光镜下观察不到棕褐色椭圆形的运动终板:而膈肌局部梭曼中毒1和10min后,全身给予HI-6对局部中毒膈肌胆碱酯酶有明显的重活化作用,在光镜下可见到肌纤维上又出现了棕褐色椭圆形的运动终板。结论HI-6全身给药对早期膈肌局部中毒的神经运动终板胆碱酯酶有重活化作用。

Abstract:
Objective To study the reactivation of HI-6 on diaphragm eholinesterase of rats with local soman intoxication. Methods Rats were anesthetized and fixed, and diaphragn was exposed by cutting the abdominal cavity along the ventral midline. After rats' diaphragms were locally intoxicated with soman within 1 and 10 min, HI-6 was administrated locally or systemically. The rats which were administrated drug were sacrificed after 24 and 72 h. The toxic parts of the diaphragms were removed, and the activity of diaphragms' end-plate AChE was showed by sulfur enzyme choline to observe the reactivation of AChE by HI-6. Results There was no effect on the diaphragm cholinesterase and no tan oval motor end plate observed through the light microscope within 24 and 72 h when HI-6 was only used locally to the diaphragm, no matter within 1 or 10 min. But there was reactivation and tan oval motor end plate observed when HI-6 was used just systemically in the same method. Conclusion HI- 6 administrated systemically has reactivation to the local toxic diaphragm motor end plate cholinesterase.
