急性出血坏死性胰腺炎大鼠循环内皮细胞与胰腺病理损伤的关系

摘    要:
目的:探讨急性出血坏死性胰腺炎大鼠循环内皮细胞、血清淀粉酶与胰腺病理损伤的关系。方法:采用胆胰管逆行注射5%牛磺胆酸钠制备急性出血坏死性胰腺炎的大鼠模型,于造模成功后3 h、6 h、12 h处死并取血,用流式细胞仪检测循环内皮细胞,用全自动生化分析仪检测查淀粉酶活性,取胰腺病理评分,进行血淀粉酶与病理评分、脱落细胞与病理评分之间的相关性分析。结果:造模成功后3 h血清淀粉酶为(5431.3±758.5)U/L,病理评分为3.3±0.8,脱落细胞计数为(35.4±3.1)%;6 h血清淀粉酶为(6450.5±669.8)U/L,病理评分为4.7±0.8,脱落细胞计数为(43.3±2.7)%;12 h血清淀粉酶为(7934.2±667.9)U/L,病理评分,6.3±0.9,脱落细胞计数为(54.8±3.8)%。血清淀粉酶与病理损伤评分之间的相关系数r=0.351,P〈0.01,关系式为Y=292.7X+5098,循环内皮细胞与病理损伤评分的相关系数r=0.842,P〈0.01,关系方程为Y=21.227+4.837X。结论:血清淀粉酶及血循环内脱落内皮细胞数量与急性出血坏死性胰腺炎时机体的损伤程度均呈正相关,但后者的相关性更强,更能反映多脏器损伤的程度。

Relationship between Rat Circulating Endothelial Cells and Pancreatic Pathology Injury in Acute Hemorrhagic Necrotizing Pancreatitis

Abstract:
Objective To explore the relationship between rat circulating endothelial cells, serum amylase and pancreatic pathology injury in acute hemorrhagic necrotizing pancreatitis. Methods The acute hemorrhagic necrotizing pancreatitis models in rats were established by retrograding cholangiopancreatography injection of5% sodium taurocholate. After the models were manufactured successfully, the rats were executed at 3 h, 6 h,12 h time points and the blood samples collected, the circulating endothelial cells were detected by flow cytometry, the activity of the amylase was checked by automatic biochemical analyzery. Then the correlation between pancreas pathology score and amylase levels was analyzed. Results At 3 hours after successful modeling, the serum amylase was(5431.3 ±758.5)U/L, the pathological score was 3.3±0.8, the circulating endothelial cell count was 35.4±3.1%; and at 6 hours after successful modeling, the serum amylase was(6450.5±669.8)U/L,the pathological score was 6.3±0.9, the circulating endothelial cell count was 43.3±2.7; at 12 hours after successful modeling, the serum amylase was(7934.2±667.9)U/L, the pathological score was 3.3±0.8, and the circulating endothelial cell count was 54.8±3.8%. The correlation coefficient between serum amylase and pathological injury score was r=0.351, P〈0.01 the relationship equation was Y=292.7X+5098, the correlation coefficient between circulating endothelial cells and pathological injury score was r=0.842, P〈0.01 the relationship equation was Y=21.227+4.837 x. Conclusion The amylase levels and the number of circulating endothelial exfoliated cells are positively correlated with the extent of damage when the body suffers in acute hemorrhagic necrotizing pancreatitis,but the latter is more relevant that can reflect the degree of multi-organ damage better.
