肺鳞癌、腺癌中多药耐药基因相关蛋白的表达及意义

摘    要:目的 检测P-糖蛋白(P-gp)、肺癌耐药蛋白(LRP)、多药耐药相关蛋白(MRP)在肺鳞癌、腺癌组织中的表达,以探讨其临床意义。方法 采用S-P免疫组织化学方法,检测105例术前未做化疗的肺鳞癌、腺癌组织中P-gp,LRP,MRP的表达水平。结果 P-gp在肺鳞癌,腺癌组织中表达阳性率为腺癌78.26%,鳞癌47.46%(P〈0.05);LRP表达阳性率为腺癌89.13%,鳞癌44.07%(P〈0.01);MRP在不同癌组织中的表达,差异无显著性(P〉0.05)。在腺癌、鳞癌组织中同时有两种或两种以上耐药基因产物表达阳性率分别为:89.13%,49.15%,两种类型问比较差异有显著性(P〈0.01)。结论 肺鳞癌,腺癌存在不同程度的耐药性,检测P-gp,LRP,MRP的协同表达对于指导临床化疗方案的实施有重要意义。

Abstract:Objective To investigate the expression level and its clinical significance of multidrug resistance(MDR) markers(P-gp,LRP and MRP) in lung squamous carcinoma and adenocarcinoma tissues.Methods The expression of lung cancer resistance protein(LRP),multidrug resistance protein(MRP) and P-glycoprotein(P-gp) of MDR markers were detected in 105 cases of lung cancer tissues by streptavidin-biotin-peroxidase(S-P) immunohistochemistry technique.Results There was overexpression of P-gp in the squamous carcinoma and adenocarcinoma tissues and the expression of P-gp in the adenocarcinoma(78.26%)was higher than that in the squamous cell carcinoma(47.46%)(P<0.05).The expression of LRP in the adenocarcinoma(89.13%)was higher than that in the squamous carcinoma (44.07%)of lung(P<0.01).No significance of difference was observed among the expression of MRP in different types of lung cancer(P>0.05).Two kinds or more of MDR markers expression in the adenocarcinoma,squamous carcinoma were 89.13%,49.15%,respectively(P<0.01).Conclusion It has different degree overexpression of multidrug resistant markers in squamous pulmonary carcinoma and adenocarcinoma tissues.Combined detection of P-gp,LRP and MRP may be useful for guiding the clinical chemotherapy of lung carcinoma.
