摘    要:目的:探讨促红细胞生成素(erythropoietin,EPO)对新生儿窒息后血清诱导的人近曲肾小管上皮细胞(HK-2)Omi/HtrA2、caspase-3表达的影响。方法:以HK-2为研究对象,分为对照组、窒息组、EPO干预组、EPO+5-HD(5-Hydroxydecanoic acid sodium salt,5-羟葵酸盐)干预组。以200 ml/L窒息后24 h血清作为攻击浓度。观察各组细胞形态,采用免疫组化法检测细胞Omi/HtrA2、caspase-3的表达。结果:与对照组相比,窒息组细胞形态变化显著,Omi/HtrA2、caspase-3表达明显增加;而与窒息组比较,EPO组细胞形态明显改善,Omi/HtrA2、caspase-3表达明显减少;加入5-HD后,EPO的保护作用被抑制。各组差异具有统计学意义(P<0.05)。结论:EPO可减少新生儿窒息后血清诱导的HK-2细胞内Omi/HtrA2、caspase-3表达。

Effect of erythropoietin on expression levels of Omi/HtrA2 and caspase-3 in renal tubular cells induced by serum of asphyxia neonatorum
Abstract:Objective:To explore the effect of erythropoietin(EPO) on expression levels of Omi/HtrA2 and caspase-3 in human renal tubular cells induced by serum of asphyxia neonatorum.Methods:Human renal proximal tubular cells were used as target cells and then divided into control group,asphyxia group and EPO group,EPO plus 5-hydroxydecanoic acid(5-HD) group.The attacking concentration of serum was 200 ml/L.The changes of cellular morphology were observed,the expression levels of Omi/HtrA2 and caspase-3 were detected by immunohistochemical staining.Results:Compared with control group,the changes of cellular morphology in asphyxia group were obvious,the expression levels of Omi/HtrA2 and caspase-3 increased significantly;compared with asphyxia group,cellular morphology in EPO group improved obviously,the expression levels of Omi/HtrA2 and caspase-3 decreased significantly;the protective effect of EPO was inhibited by 5-HD.There was significant difference between different groups(P<0.05).Conclusion:EPO can relieve the expression levels of Omi/HtrA2 and caspase-3 in human renal tubular cells induced by serum of asphyxia neonatorum.
