摘    要:目的研究BRCA2(乳腺癌易感基因2)在肺鳞癌中的表达情况,以筛选肺鳞癌分子标志物。方法利用cDNA微阵列技术研究6个分期为Ⅰ期的手术切除鳞癌标本的基因表达谱,重点分析BRCA2的表达情况,并利用免疫组化方法测定其蛋白表达水平。结果BRCA2基因在全部6个肺鳞癌标本中均呈下调表达,免疫组化结果显示BRCA2蛋白在20个肺鳞癌标本中表达较低(与20例正常肺组织比较),与cDNA微阵列技术检测结果相符。结论cDNA微阵列技术结合免疫组化可用于肺鳞癌分子标志物的筛选,BRCA2可初步作为肺鳞癌的侯选分子标志物。

Abstract:Objective To investigate the expression of breast cancer susceptibility gene 2(BRCA2) in lung squamous cell carcinoma to find out the molecular marker in lung squamous cell carcinoma.Methods Gene expression profiles of 6 surgical specimens of lung squamous cell carcinomas of stage I were identified by using cDNA microarrays.The expression of BRCA2 expression was analyzed by using microarrays and immunochemical assay.Results The expression of BRCA2 was down-regulated in all six specimens.The results of immunhistochemical analysis showed that BRCA2 expression was significantly lower in 20 specimen of squamous cell carcinoma than that in 20 normal lung tissue specimen.Conclusions BRCA2 can be considered as one of the molecular markers in lung squamous cell carcinoma.The cDNA microarray combined with immunochemical assays can be used in the screening of the molecular markers in lung squamous cell carcinoma.
