摘    要:目的:研究失血性休克时细胞线粒体呼吸功能变化及防治方法。方法:将24只Wistar大鼠随机均分为假手术组、模型组和治疗组,模型组和治疗组颈动脉快速放血造成失血性休克模型,维持1.5h后复苏。假手术组动物除不放血及复苏外其他操作同模型组。治疗组于实验前2d开始及放血前4h各给予静脉注射中药益气活血解毒汤1mL/kg干预治疗。于复苏后4h处死动物,测定肝匀浆内毒素和线粒体呼吸功能。结果:与假手术组比较,模型组肝脏内毒素水平明显升高(P<0.01),而Ⅲ态呼吸耗氧速率(RS3)和呼吸控制率(RCR)下降显著(P<0.01)。与模型组比较,治疗组肝脏内毒素的升高及RS3、RCR的降低明显减轻(P<0.05)。结论:大鼠失血性休克复苏后肝线粒体呼吸功能严重受损,以RS3下降为主,中药益气活血解毒汤治疗可明显改善线粒体功能。

Abstract:Objective: To study the effect of Chinese herbs on mitochondrial respiratory function in hemorrhagic shock in wistar rats. Methods: Twenty-four Wistar rats were randomly divided into three groups, model group, false operation group, and Chinese herbs treatment group. To establish a hemorrhagic shock model, Wistar rats were drawn blood from carotid artery rapidly and were resuscitated 1.5 hours later. The Yiqihuoxuejiedutang was given intravenously in treatment group. All rats were killed 4 hours after resuscitation, and the liver was dissected to determinate the levels of endotoxin and mitochondrial respiratory function. Results: Compared with false operation group , the level of liver endotoxin in model group was higher, but the respiratory stage III(RS3)and respiratory control ratio(RCR)were decreased significantly(P < 0.01). However, little changes were observed between treatment and model group(P < 0.05). Conclusion: The mitochondrial respiratory function of rat liver was insulted severely during hemorrhagic shock, mainly with the significant decrease of RS3. The treatment of Chinese herbs (Yiqihuoxuejiedutang) can improve the mitochondrial respiratory function significantly in hemorrhagic shock in rats.
