CD71和HbF用于流式细胞仪分选母血中的胎儿细胞

目的:比较CD71单染、HbF单染和CD71、HbF双染对单个核细胞的标记情况,同时探讨孕妇外周血中胎儿有核红细胞的含量与孕周的关系。方法:选用红细胞特异性抗体CD71、HbF对正常孕妇外周血和胎儿脐血单个核细胞进行标记,并结合流式细胞仪对有核红细胞进行分选。结果:用CD71和HbF进行双染色标记,正常孕妇和脐血单个核细胞的阳性标记率明显低于CD71和HbF单染色标记(t检验P<0.01)。孕早中期胎儿NRBC比例随孕周的增加呈上升趋势,至21~24周比例最大,约为1.92±0.84×10-2。结论:采用CD71和HbF双染色标记,可提高NRBC的特异性标记和分选纯度,分选胎儿细胞的最佳时间应在孕21~24周。

Sorting fetal cell from maternal blood using CD71 and HbF double-staining method flow cytometry

Objective: To compare mononuclear cells labeled by CD71 or HbF single-staining method with CD71 and HbF double-staining one, and study the relationship between the proportion of fetal cells and gestational age.Methods:Mononuclear cells in umbilical blood and peripheral blood in normal pregnant women were marked by erythrocyte specific antibody CD71 and fetal specific antibody HbF.Furthermore,NRBCs were sorted with flow cytometryResults:Compared double-staining method with single-staining method,the positive labeling rates of mononuclear cells in all cases were significantly different.(t-test,P<0.01).The proportion of NRBCs sorted by flow cytometry was increased until 21~24 week's gestation(1.92±0.84×10-2).Conclusion:CD71 and HbF double-staining method can improve NRBCs specific labeled and sorting purity and make advantage for the next prenatal diagnosis.The best time of sorting fetal NRBCs in maternal blood is at the 21~24 week.
