摘 要: | 目的 通过研究苯那普利对糖尿病大鼠肾脏氧化应激及羰基应激的影响,探索它的肾保护机制.方法 大鼠分为正常对照组(A组)、未处理糖尿病组(B组)和苯那普利治疗组(C组).12周后测定各组大鼠肾重/体重比、血糖、糖化血红蛋白(HbAle) 24 h尿蛋白定量,并分析各组大鼠肾组织丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、总巯基及羰基水平.结果 B组大鼠肾重/体重比、24 h尿蛋白定量、肾组织MDA及羰基水平均明显高于A组,SOD、GSH-PX及总巯基低于A组.与B组相比,C组大鼠肾重/体重比、24 h尿蛋白定量、肾组织MDA及羰基水平下降,GSH-PX升高.结论 苯那普利可抑制糖尿病大鼠肾脏肥大,并降低尿蛋白排泄率,这种肾保护作用可能与其改善氧化应激及羰基应激有关.

Effect of benazepril on oxidative stress and carbonyl stress in renal tissues of diabetic rats |

Abstract: | Objective To investigate the effects of Benazepril on oxidative and carbonyl stress in diabetic rat kidneys, and to explore the renoprotective mechanisms in diabetic rats. Methods Rats were randomly divided into three sub-groups, including, normal control (group A) , diabetic control (group B) and diabetic with Benazepril (group C). Blood glucose, HbAlC, the ratio of kidney to body weight and urinary protein excretion were measured. Malonyldialdehyde (MDA) level, superoxide dismutase (SOD) activity, glutathione peroxidase (CSH-Px) activity, total sulfhydryl group and protein carbonyl levels in renal tissue were also determined. Result Compared to group A, the ratio of kidney to body weight, urinary protein excretion, MDA and protein carbonyl levels in group B were significantly higher, while SOD,GSH-Px activities and total sulfhydryl group level were significantly lower. The ratio of kidney to body weight, urinary protein excretion, MDA and protein carbonyl level in group C were significantly lower than those in group B. GSH-Px activity in group C were higher than that in group B. Conclusion Benazepril can prevent renal hypertrophy and decrease urinary protein excretion in diabetic rats. The renoprotective effects of this drug may be related to the ability to inhibit oxidative and carbonyl stress. |
