血瘀证与血管内皮细胞内游离钙浓度关系的实验研究 |

摘 要: | 目的:研究血瘀证与血管内皮细胞内游离钙浓度的关系,观察不同复方(活血化瘀复方、益气活血复方、理气活血复方)对血内皮细胞内游离钙浓度的影响。方法:以大鼠为研究对象,给予高胆固醇和高脂饲料喂养建立血瘀证动物模型,分为6组,观察组分别给予硝苯地平、活血化瘀复方、益气活血复方、理气活血复方。下腔静脉取血,测血清钙、血清总胆固醇和三酰甘油,Fura-2/Am标记,荧光测定血管内皮细胞内游离钙浓度。结果:①造模组与对照比较,血清钙明显升高(P<0.05)。②所有复方均具有钙通道阻断剂样作用,能增加细胞外钙浓度,与造模组比较差异显著(P<0.05)。③造模组与对照组比较,胞内游离钙浓度明显升高(P<0.05),益气活血复方能使造模运动血管内皮细胞内游离钙浓度降低,与造模组比较差异显著(P<0.05)。结论:①血瘀证血管内皮细胞内游离钙浓度升高,血瘀证的形成可能与血管内皮细胞的屏障功能下降有关。②益气活血复方能降低胞内游离钙浓度,具有血管内皮保护作用。

EXPERIMENTAL STUDY ON CONNECTION OF BOLLD STASIS SYNDROME AND [Ca~(2 )]_i IN VASCULAR ENDOTHELIAL CELL |

Abstract: | Obuective:To research the connection of blood stasis syndrome and Ca\+\{2+\}]\-i in vascular endothelial cell(VEC) and observe the influence of different compound recipes(Huoxue Huayu,Yiqi Huoxue,Liqi Huoxue) on Ca\+\{2+\}]\-i in VEC.Method:The rat models were established successfully through feeding them high-cholesterol and high-fat foods.All rats were divided into 6 groups.The model groups were treated by different medinines which were nifedipine,the recipe of Huoxue Huayu,the recipe of Yiqi Huoxue and the recipe of Liqi Huoxue.The blood was taken,and sero-Ca\+\{2+\}],TC],TG]were estimated.Ca\+\{2+\}]\-i was fluoremetried by means of Fura-2/AM.Result:①Sero-Ca\+\{2+\}] in model groups was higher than that in the control group(P<0 05).②There was significant differece in raising Ca\+\{2+\}]\-i in serum between the Chinese medicine groups and the nifedipine group (P<0.05).③Ca\+\{2+\}]\-i in VEC of the model group was higher than that of the control group.The recipe of Yiqi Huoxue could make Ca\+\{2+\}]\-i even lower than the others treated by the other recipes.There was significant difference comparing to model group(P<0.05).Conclusion:①When the bolld stasis syndrome happened,Ca\+\{2+\}]\-i in VEC of rats was higher than that of the normal rats.It also showed that the occurrence of blood stasis was probably due to the reduce of endothelial cell's barrier function.②The recipe of Yiqi Huoxue could reduce Ca\+\{2+\}]\-i ,so it had the protective effection on the endothelial cell. |
