应用环孢素A治疗异种肝细胞移植的排斥反应

摘    要:目的 探讨应用环孢素A(CsA)治疗异种肝细胞移植排斥反应的疗效及其排斥机制。方法 将豚鼠肝细胞植入D-氨基半乳糖诱导的肝衰大鼠脾内,分CsA治疗组和单纯移植组。观察2周生存率及脾病理组织学检查;酶联免疫双抗体夹心法(Sandwich-ELISA)测定大鼠血清白细胞介素(IL)-2的浓度。结果 2周生存率比较CsA治疗组(78.6%)与单纯移植组(80.0%)相比差异无显著性(P>0.05)。应用CsA可减缓炎症细胞浸润。血清IL-2浓度检测正常大鼠为(28.7±7.0)ng/L,移植后各组血清IL-2浓度变化不大,直至移植后第7天,CsA治疗组为(28.5±6.0)ng/L、单纯移植组为(31.5±8.0)ng/L。3组间差异均无显著性(P>0.05)。结论 在异种肝细胞脾内移植中,细胞免疫发生的较迟和/或较弱。单独应用CsA治疗异种肝细胞脾内移植引发的排斥反应其疗效不佳。

Abstract:Objective To investigate the immunosuppressive effect of cyclosporine A (CsA) on xenogeneic hepatocyte transplantation (XHT) and the rejection mechanisms.  Methods The rats with hepatic failure induced by D-GI were divided into 2 groups: CsA-treated (10 mg/kg per day by intraperitoneal injection) group and CsA-untreated group. The hepatocytes of guinea pig were transplanted into the spleen of the rats. The splenic pathohistological examination in the recipients was performed and the 2-week survival rate in the 2 groups observed. Sandwich-ELISA method was used to determine the serum levels of interleukin-2 in the rats.  Results The survival rate in the CsA-treated group (78.6 %) and CsA-untreated group (80.0 %) had no significant difference (P>0.05). But CsA could attenuate the infiltration of inflammatory cells. No significant difference was found in the levels of serum IL-2 of the rats among the normal (28.7±7.0) ng/L, CsA-treated(28.5±6.0) ng/L and CsA-untreated groups [(31.5±8.0) ng/L, P>0.05].  Conclusion  Cellular immune response would occur later and would be weaker in xenogenic hepatocellular transplantation. CsA has no significant therapeutic effects on XHT.
