实验性脑出血血肿周围组织的细胞凋亡

摘    要:
目的 观察实验性脑出血血肿周围组织的细胞凋亡情况。方法 采用犬脑内注入非肝素化的自体动脉血建立实验性脑出血动物模型。分别在术后6h、24h、48h、72h和7d处死动物,取脑,制成单细胞悬液,流式细胞仪检测凋亡细胞的百分率。结果 注血后实验动物有类似于自发性脑出血的行为变化,大体和光镜下符合脑出血后的病理改变。血肿形成后上述各时间点凋亡细胞所占百分率分别为:9.69%、15.47%、25.67%、29.06%和37.46%。结论 犬脑内注入非肝素化的自体动脉血可建立实验性脑出血的动物模型。血肿形成后6h即可检测到凋亡细胞的存在,血肿周围组织中凋亡细胞所占的比例在7d内随时间延长逐渐增加。

Abstract:
Objective Apoptosis following experimental intracerebral hemorrhage(ICH) was detected by flow cytometry. Methods The experimental ICH model was established by injection of non-heparinized arterial blood into canine brain. The ICH dogs were executed respectively at 6h,24h,48h,72h and 7d after operation. The percents of apoptosis were detected by flow cytometry. Results The behavioral and pathological changes of ICH dogs were similar to those of spontaneous ICH. The percents of apoptosis of the time points mentioned above were 9.69% ,15.47%,25.67%,29.06% and 37.46% respectively. Conclusion The experimental ICH model can be established by injection of non-heparinized arterial blood into canine brain. Apoptosis may be detected 6h after formation of hematoma and the percents of apoptosis increase gradually as time passing on.
