利血平致大鼠脾虚模型尿D-木糖排泄率与肠黏膜三磷酸腺苷水平的研究

摘    要:[目的]观察利血平致脾虚大鼠的病理改变情况,为在该模型上观察益气健脾方药的作用提供参考。[方法]模型组以利血平注射造模15 d,停药后再观察30 d,实验周期共45 d;另设正常组观察45 d。观察大鼠一般状态、体重、尿D-木糖排泄率、小肠黏膜三磷酸腺苷(ATP)水平、胃肠黏膜病理组织学、脏器系数等指标。[结果]模型大鼠体重和尿D-木糖排泄率与正常组比较差异有统计学意义(P〈0.01),持续至45 d;小肠黏膜ATP水平比正常组略为降低;尿D-木糖排泄率改变与小肠黏膜ATP水平改变呈正相关;胃肠黏膜病理组织学检查未见明显改变;肝和胸腺脏器系数降低。[结论]利血平所致脾虚大鼠病变状态可持续较长时间,与益气健脾方药缓慢起效的特点相符;有肠道消化吸收功能降低的改变,并伴随肠黏膜ATP水平的降低。该动物模型可用于观察益气健脾方药改善消化吸收功能等作用。

Abstract:[Objective]To observe the pathological changes and D-xylose excretion rate, ATP content in the small intestines mucosa membranes of rats with spleen deficiency induced by reserpine injection to provide reference for study on formula and herbs of strengthening spleen. [Methods]Spleen deficiency rat model was made by Reserpine injection for 15 days, and then continue observing 30 days, comparing with normal group. General conditions, body weight, D-xylose excretion rate, ATP content and histopathological study in mucosa were measured, [Results] The body weight and D-xylose excretion rate decreased after reserpine injection (P〈0. 01 ,vs. control group). The mucosal ATP content of model rats appreciably decreased, there was obvious positive correlation between change of urine's D-xylose excretion rate and mucosa ATP content. No obviously change was detected in histopathologic examination. The organ coefficient of liver and thymus were lower than that of normal rats. [Conclusion ]Pathogenic state of spleen deficiency rat caused by reserpine injection can last a long period, which is suitable for the characteristic of Chinese herb and formula for strengthening spleen. The model rats have dysfunction of digesting and absorbing, associated with mucosal ATP contents decreasing. So it can be used for studying Chinese herb and formula for strengthening spleen.
