| 摘 要： | 目的验证大鼠腹腔巨噬细胞环氧合酶-2抑制剂体外筛选模型的可靠性。方法采用放免法测定用脂多糖刺激后的大鼠腹腔巨噬细胞前列腺素-2的产量,运用Western印迹法检测处理后的大鼠腹腔巨噬细胞COX-2和COX-1的表达。结果加入0.5μg.L-1浓度的LPS刺激6 h后,大鼠腹腔巨噬细胞前列腺素-2的产量最高,COX-2蛋白表达量最高;加入阿司匹林后,大鼠腹腔巨噬细胞COX-1表达明显降低。结论大鼠腹腔巨噬细胞模型可用于COX-2抑制剂体外筛选。

| Abstract: | Aim To validate the dependability of screening models by culture of rat macrophages in vitro.Methods The PGE2 was detected by using radioimmunoassay in different concentration of serum,stimulating time and concentration of lipopolysaccharide（LPS）.The target protein COX-1 and COX-2 were detected by Western blotting in different scrum concentration,stimulating time and concentration of LPS.Results Macrophages produce more PGE2 after stimulated by LPS in concentration of 0.5 μg· L-1 for 6 hours,meanwhile,produced more COX-2 than others. There is not an obvious difference on COX-1 between O. 5% and 10% FCS RPIM-1640,but it decreases visibly after aspirin added to it. Conclusion The screening models can be used for screening selective COX-2 inhibitors of the antiinflammatory drugs. |
