白芍总甙对大鼠腹腔巨噬细胞化学发光的影响

摘    要:本文采用正交设计L_(25)~((5)4)对影响大鼠腹腔巨噬细胞(PMφ)鲁米诺依赖的化学发光(LDCL)实验因素—鲁米诺浓度、细胞浓度、酵母多糖浓度、酵母多糖调理时间等进行了探讨,表明其最佳反应组合水平为:100μmol/L—5×10~6/ml—10g/L—45min大鼠腹腔Mφ在有或无白芍总甙(TGP)的条件下体外培养12h,用上述各因素的亚适水平进行LDCL测定,结果表明,TGP在0.09—2.25mg/L范围内可浓度依赖性地增加Mφ的LDCL强度,当TGP浓度增至11.25mg/L时,CL降低,量效曲线呈钟罩形趋势,提示该药对LDCL的影响有双向作用。

Abstract:Influences of the concentrations of luminol, zymosan & cells and opsonizing time of zymosan on luminol-dependent chemiluminescence( LDCL ) of peritoneal macrophages ( PMφ) in rats were investigated by the method of orthogonal analysis. The result showed that the best levels were 100μmol/L of luminol, 10 mg/ml of zymosan, 5× 10~6/ml of cells and 45 min for opsonizing time. The LDCL of PMφ was enhanced by incubation with TGP for 12 h in a concentration-dependent manner in the range of 0.09-2.25 μg/ml of TGP, but LDCL was diminished with the concentration of 11.25 μg/ml of TGP . The concentration-effect curve seems to be bell-shaped. The data suggested that TGP had two-sided regulatory effects on the LDCL of PMφ in rats, depending on the concentration used.
