滋肾调冲法对诱发排卵大鼠卵巢激活素A表达的影响

摘    要:[目的]观察滋’肾调冲法对诱发排卵大鼠卵巢激活素A表达的影响,探讨该法促进卵泡发育及其排卵的分子作用机制[方法]将未成年雌性大鼠随机分成中药高、低剂量组、阳性对照组和模型组,分别给予功血宁Ⅱ号方、安坤赞育丸和生理盐水14d后,各组大鼠每只颈部皮下注射妊马血清促性腺激素(PMSG)50IU,48h后每组随机选取10只大鼠处死后摘取卵巢,此为卵泡发育期模型。每组各余下10只大鼠,每只颈部皮下注射人绒毛膜促性腺激素(HCG)30IU,12h后各组所有大鼠处死后摘取卵巢,此为排卵前期模型。常规制备卵巢切片,免疫组化法检测各组卵巢不同发育阶段卵泡激活素A的表达。[结果]在卵泡发育期和排卵前期,功血宁Ⅱ号方激活素A的阳性面积、阳性面积率、积分光密度以及平均光密度均显著高于模型组(P〈0.05或P〈0.01),高剂量组的作用较低剂量组和阳性对照组显著。各组排卵前期的上述指标均明显高于各组的相应卵泡发育期(P〈0.05或P〈0.01)。[结论]滋肾调冲法促进卵泡生长发育及其排卵的作用机制与其提高卵巢激活素A的表达有关。

Effect of zishen tiaochong method on the expression of ovaries activin a in rats with induced ovulation

Abstract:[Objective] To observe the effect of Zishen Tiaochong(ZSTC) method on the expression of ovaries activin A in rats with induced ovulation and explore the molecular action mechanism on promoting follicular development and ovulation. [Methods] The female rats were randomly divided into the high and low dosage of TCM group, positive control group and model group and were individually administrated to Gongxuening H ,Ankunzanyu pills and physiologic saline in fourteen days. To establish the model of follicular development stage by injecting PMSG(50 IU) and establish the model of preovulatory phase by injecting HCG (30 IU). To prepare the ovaries sections and determine the expression of ovaries activin A in rats with induced ovulation in different developmental stage by using immunohistochemical method. [Reslflts] The ovaries activin A of Gongxuening 11 group was obviously higher than that of model group on the masccline area, the rate of masccline area, photodensity value and average optical of follicular development stage and preovulatory phase (P〈0.05 or P〈0.01). The effect of high dosage group were obviously higher than that of low dosage and positive control group. The above indexes of follicular development stage was obviously higher than that of preovulatory phase in rats of every group (P〈0.05 or P〈0.01). [Conclusion] Zishen Tiaochong method may promote growth and development of ovarian follicle and ovulation and its effect mechanism may have a related with increasing the expression of ovaries activin A.
