几株抗血吸虫单克隆抗体细胞株的建立与特性鉴定

摘    要:
为了对血吸虫病的免疫诊断和免疫预防提供有价值的单克隆抗体,通过2次骨髓瘤细胞与感染日本血吸虫的小鼠脾细胞间的融合试验和多次筛选以及克隆化,建立了4株分泌单克隆抗体的细胞株。其中2株的靶抗原为血吸虫成虫肠道;1株为成虫表面膜;另一株为成虫的肌细胞或纤维细胞。免疫球蛋白亚类鉴定表明前3株均为IgG_1。除了SM 48-2对虫卵呈弱反应外,未见对血吸虫虫卵、尾蚴、童虫有阳性反应;亦未见对曼氏血吸虫、肺吸虫、华枝睾吸虫、疟原虫及利什曼原虫有阳性反应。这几株细胞已在体外培养下稳定地分泌特异性抗体达6个月之久。


PREPARATION AND CHARACTERIZATION OF SEVERAL MONOCLONAL ANTIBODY CELL LINES AGAINST SCHISTOSOMA JAPONICUM
Abstract:
In order to supply valuable monoclonal antibodies for immunodiagnosis and immune prevention, through two fusions between myeloma cells and spleen cells of mice infected with Schistosoma japonicum and a lot of screenings and clonings, 4 monoclonal antibody cell lines have been established. Of these, two lines showed against digestive tract of the adult worm, one showed against surface membrane and another against muscle or fiber cells. Immunoglobulin subclasses of front three lines were determined to be IgG1. Except monoclonal antibody No 48-2 showed weak positive reaction to egg, no positive reaction was found to egg, cercariae and shistosomule. Also as no reaction was found to Schistosoma mansoni, Paragonimus, Clonorchis, Malaria and Leishmanis. These cell lines were stable to secrete monoclonal antibodies against Schistosoma japonicum in tissue culture near'ly for 6 months.
