全反式维甲酸对人牙周膜细胞向成骨分化的影响

摘    要:目的检测全反式维甲酸(AT-RA)对人牙周膜细胞(hPDLCs)向成骨分化的影响。方法原代培养hPDLCs,用噻唑盐(MTT)方法检测AT-RA作用于细胞hPDLCs及对其增殖的影响,用酶动力方法检测对其碱性磷酸酶(ALP)活性的影响,用茜素红染色方法检测对其形成钙盐沉积的影响。结果 AT-RA不影响hPDLCs的增殖活性,但能增强hPDLCs的ALP活性,并明显促进钙盐沉积。结论 AT-RA可以促进hPDLCs向成骨分化。

Abstract:【Objective】 To detect the effect of all-trans retinoic acid on osteogenic differentiation of human periodontal ligament cells(hPDLCs).【Methods】 hPDLCs was cultured in vitro in DMEM containing AT-RA or not.The proliferation of the hPDLSCs was measured by MTT colorimetric assay.The alkaline phosphatase activity was determined by enzyme kinetic method.The calcium deposit was determined by alizarin red staining.【Results】 The proliferation of the hPDLCs was not affected by AT-RA.However,the alkaline phosphatase activity and the calcium deposit were obviously raised by AT-RA.【Conclusions】 AT-RA can promote the osteogenic differentiation of hPDLSCs.
