摘    要:
目的 观察蝎毒抗癌多肽(APBMV)对小鼠肝癌(H22)实体瘤及瘤细胞的抑制作用。方法 应用MTT法检测APBMV对H22瘤细胞的毒性作用;通过小鼠H22实体瘤模型,观察APBMV对肿瘤生长的抑制作用及对小鼠外周血白细胞(WBC)数和脾脏指数(SI)的影响。结果 APBMV对H22细胞具有一定的毒性作用,但剂量效应关系不明显(P>0.05)。APBMV能明显抑制小鼠H22肿瘤的生长,抑制率最高可达40.30%(P<0.01),带瘤小鼠外周血WBC和SI无明显变化或略高于对照组,而阳性对照5-Fu组小鼠WBC和SI均明显下降(P<0.01)。结论 APBMV是东亚钳蝎蝎毒中的1种低毒有效的抗肿瘤成分,能抑制小鼠肝癌的生长。

Abstract:
Objective To observe the inhibition effect of antineoplastic polypeptide from Buthus martensii venom(APBMV) on H 22 solid neoplasm and tumor cells.Methods MTT colorimetric method was used to measure the toxicity of APBMV on H 22 tumor cell;Applying H 22 bearing mice model,we observed the growth inhibitory effect of APBMV on tumor,also the influence on peripheral white blood cell(WBC) number and spleen index(SI) were investigated.Results The ability to metabolize MTT of H 22 cells was lower distinctively after the cells were treated by APBMV,wherase dose response relationship was not clear( P >0.05).Tumorgrowth of H 22 bearing mice was inhibited by APBMV evidently,the growth inhibiting rate was 40.30%( P <0.01).After treatment by APBMV,peripheral WBC number and SI of H 22 bearing mice were not changed markedly or slightly higher compared with control group.In positive control 5 fluorouracil group,WBC number and SI of H 22 bearing mice were all reduced distinctly( P < 0.01 ).Conclusion APBMV is one of effective and low toxicity antineoplastic components in the venom of Buthus Martensii Scorpion.It possesses growth inhibiting ability on the mice liver neoplasm.
