Abstract:OBJECTIVE To investigate the effects of Periplaneta Americana extract(PA-B) on oxidative stress and hepatic fibrosis index induced by CCl4 in rats.METHODS The rat hepatic fibrosis model was established by injected intraperitoneally with 40% CCl4 oil solution.Forty-eight rats were randomly divided into normal control group, hepatic fibrosis model group, colchicine group, low-dose PA-B group, medium-dose PA-B group and high-dose PA-B group, with 8 rats in each group.After 4 weeks of treatment, blood was collected under anesthesia, fasting without water for 12 h.Serum levels of ALT, AST, HA, LN, PC-Ⅲ, Col-Ⅳ, GSH-Px, CAT, T-SOD and MDA were determined.Pathological changes of liver tissues were observed by HE staining and Masson staining.RT-qPCR was used to detect mRNA expressions of Nrf2, NQO1, HO-1, Keap1 and CAT in liver tissues, and the protein expressions of Nrf2, NQO1, HO-1 and Keap1 in liver tissues were detected by Western Blot.RESULTS PA-B could significantly decrease the serum levels of AST, ALT, HA, LN, PC-Ⅲ, Col-Ⅳ and MDA, and increase the levels of GSH-Px, CAT and T-SOD(P<0.01).CAT mRNA expression was significantly up-regulated(P<0.01).The mRNA and protein expressions of Nrf2, NQO1 and HO-1 were significantly up-regulated(P<0.01).It significantly down-regulated Keap1 mRNA and protein expression(P<0.01).CONCLUSION PA-B can alleviate CCl4-induced hepatic fibrosis, and its mechanism may be related to the regulation ofoxidative stress balance through Nrf2 signaling pathway.
