Survivin和PLK1在急性白血病中的表达及临床意义

目的：探讨保罗样激酶1（polo样激酶 PLK1）和抗凋亡蛋白（Survivin）在急性白血病（AL）中的表达水平及临床意义。方法：应用免疫细胞化学技术（ICC）检测54例AL初治患者骨髓细胞中PLK1和抗凋亡蛋白Survivin的表达水平，经过标准化化疗方案化疗后分为缓解组、未缓解组。并取19例健康体检成人（同期闻在本院确诊的非血液病患者）骨髓标本作为对照组。结果：PLK1和Survivin在初治急性髓细胞性白血病（AML）和急性淋巴细胞白血病（AL）患者骨髓细胞中的表达均显著高于正常对照组和完全缓解组，其显著性差异存在统计学意义（P〈0．05）；PLK1和Survivin高表达组CR率低于低表达组，差异有显著性（P〈0．05）；二者的表达呈正相关。结论：PLK1和Survivin的过表达对急性白血病的发生和发展有着不同程度地影响，有希望成为急性白血病预后判断的监测指标。PLK1和Survivin表达呈正相关，提示PLK1的过表达可能促进Survivin的表达，增强Survivin的抗凋亡活性。

Expression of PLK1 and Survivin in acute Leukemia and their clinical significant

Objective：To study the expression of PLK1 and Survivin in acute Leukemia, and to value their clinical significance. Methods：54 patients with uncured acute Leukemia were determined by using immunocy- tochemistry （ICC）. After treatment, 30 patients completely remitted（CR）. The serum levels of PLK1 and Survivin of all these groups were compared with 19 normal controls. Results： The expression of PLK1 and Survivin in uncured AML and ALL were significantly higher than those in normal control group and CR patients （P 〈 0.05）. The complete remission of PLK1 and Survivin were lower in high expression groups than those with the low expression groups. PLKi level correlated with Svrvivin positively （r = 0. 515,P ~ 0.05）. Conclusion： The expression of PLK1 and Survivin increased in AL patients compared to normal control and CR patients, which indicated that PLK1 and Survivin might play an important progression of AL. In addition, our study showed that PLK1 and Survivin expression levels were positively correlated, which suggests that up-- regulation of PLK1 enhance the expression of Survivin and the ability of Survivin to prevent apoptosis.
