雄芍汤抗大鼠免疫性肝纤维化作用研究

目的研究雄芍汤抗大鼠免疫性肝纤维化的作用。方法 84只SD大鼠随机分为对照组、模型组、扶正化瘀胶囊阳性对照组、雄芍汤预防组和雄芍汤高、低剂量(生药15.8、7.9 g/kg)组(造模后给药)。采用ip猪血清法制备大鼠肝纤维化模型,观察雄芍汤对肝纤维化大鼠血清肝功能指标丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、白蛋白(ALB)、白蛋白与球蛋白比值(A/G)和肝纤维化血清标志物透明质酸(HA)、层粘连蛋白(LN)、III型前胶原(PCIII)、IV型胶原(CIV)水平,以及肝组织病理学的影响。结果与模型组比较,雄芍汤各给药组血清ALT、AST、HA、LN、PCIII水平明显降低(P<0.05、0.01),血清ALB水平和A/G值明显升高(P<0.01);雄芍汤预防组血清CIV水平明显降低(P<0.05);所有给药组肝细胞变性和坏死程度轻微,肝组织纤维增生较少,雄芍汤高剂量组和预防组的作用尤为明显。结论雄芍汤能减轻免疫性肝纤维化大鼠的肝细胞损伤,改善肝功能,调节细胞外基质的代谢,阻止或逆转肝纤维化。

Effects of Xiongshao Decoction on immune liver fibrosis in rats

Objective To research the effects of Xiongshao Decoction(XSD) on immune liver fibrosis in rats.Methods Eighty-four SD rats were randomly divided into normal control group,model group,Fuzheng Huayu(FZHY) Capsule group,XSD prevention group,XSD high-dose and low-dose groups(15.8 and 7.9 g/kg).The hepatic fibrosis model was induced by injecting porcine serum to the abdominal cavity of rats.To observe the effects of XSD on the serum hepatic function of rats: ALT,AST,ALB,and A/G,the serum marker of hepatic fibrosis: hyaluronic acid(HA),laminin(LN),precollagen III(PCIII),and collogen IV(CIV),and hepatic tissue pathomorphology as well.Results Compared with the model group,the levels of ALT,AST,HA,LN,and PCIII in every XSD treatment group were obviously decreased(P<0.05,0.01),the levels of ALB and A/G were obviously increased(P<0.01).The CIV level of serum in XSD prevention group was significantly decreased(P<0.05) and the degrees of hepatocyte degeneration and necrosis in all treatment groups were lower,with less liver fibrosis,especially those in XSD prevention group and high-dose group.Conclusion XSD could not only relieve the degree of liver cells damage of immune hepatic fibrosis in rats,improve hepatic function of them,and regulate the extracellular matrix metabolism,but also arrest the hepatic fibrosis or make it reversed.
