痛经康口服液对痛经大鼠子宫组织中催产素受体及加压素受体mRNA的表达的影响

目的观察痛经康口服液(延胡索、当归、川芎、香附等)抗痛经作用及相关机制。方法大鼠股部皮下注射己烯雌酚,1次/d,连续11 d,其中第1天及第11天注射量为0.8 mg/只,第2天至第10天注射量为0.4 mg/只。于最后1次给药40 min后,所有大鼠按4 U/kg体质量腹腔注射缩宫素致大鼠扭体反应。采血,处死,取子宫组织采用RT-PCR方法检测大鼠子宫组织催产素受体(OTR)及加压素受体(VPR)mRNA的表达情况。结果痛经康低、中、高剂量组出现扭体反应的动物数量均较模型对照组少。田七痛经胶囊组、痛经康中、高剂量组出现扭体反应潜伏期较模型对照组延长,30 min扭体反应次数较模型对照组减少,其中痛经康中剂量组与模型对照组比较扭体反应潜伏期明显延长(P<0.01),但扭体反应次数无统计学差异。痛经康低、中、高剂量组可不同程度降低痛经大鼠子宫催产素及加压素受体mRNA表达,与模型组比较差异显著(P<0.01,P<0.05)。结论痛经康口服液具有明显的抗痛经作用,机制可能与下调痛经大鼠子宫催产素及加压素受体表达有关。

Influence of Tongjingkang Oral Liquid on the oxytocin receptor and vasopressin receptor mRNA expressions in the uteru of dysmenorrhea model rats

AIM To investigate the effect of Tongjingkang Oral Liquid(Corydalis Rhizoma,Angelicae sinensis Radix,Chuanxiong Rhizoma,Cyperi Rhizoma,etc.) on dysmenorrhea in rats and its mechanism.METHODS Rat models for dysmenorrhea were established with successively subcutaneous injection of diethylstilbestrol once a day for 11 d except control group.Rats in dysmenorrhea model group were intraperitoneal injection of oxytocin in a dose of 4 U/kg body weight in 40 minutes after the last administration.The frequency of writhing was recorded in 30 min.RT-PCR method was used to observe the oxytocin receptor(OTR) and vasopressin receptor(VPR) mRNA expressions in rat uteruine.RESULTS Compared with that of the model group,the frequency of stretching decreased in all Tongjingkang control groups.The writhing latent period was prolonged in the high dose control group and the middle dose control group of Tongjingkang and Tianqi Tongjing Capsule group,and the difference was significant in the middle dose group as compared with the model group(P<0.01).The incidence rate decreased in the high dose control group and the middle dose control group of Tongjingkang and Tianqi Tongjing Capsule group.The expressions of OTR mRNA,VPR mRNA in rat uterine tissue were significantly lower in the Tongjingkang control group than those in the model group of varying degree(P<0.01,P<0.05).CONCLUSION Tongjingkang Oral Liquid has an analgesic effect on dysmenorrhea induced by oxytocin in rats,and its mechanism may be associated with down-regulated the expressions of OTR mRNA and VPR mRNA.
