小归芍化浊解毒方对胃癌前病变大鼠PCNA及Reg基因干预作用的实验研究

目的探讨小归芍化浊解毒方(当归、白芍、藤梨根、瓜蒌、茯苓等)对实验性胃癌前病变的作用和机制。方法按随机数字表把100只大鼠随机分成为A(空白)组、B(模型)组、C(中药大剂量)组、D(中药小剂量)组、E(阳性对照)组,每组20只,除空白组外,其余各组采用N-甲基-N-硝基N-亚硝基胍(MNNG)配合饥饱失常诱导造模。造模后A组正常喂养;B组予蒸馏水灌胃;C组按30 g/kg予小归芍化浊解毒方大剂量混悬液灌胃;D组按20 g/kg予小归芍化浊解毒方小剂量混悬液灌胃;E组将胃复春混悬液灌胃。测定胃组织PCNA及RegI的表达量。结果 (1)PCNA的表达量:小归芍化浊解毒方大小剂量组与胃复春比较,能明显降低PCNA的表达水平(P<0.05)。(2)RegI的表达量:小归芍化浊解毒方大剂量组与胃复春组比较,能明显降低RegI的表达水平(P<0.05)。结论小归芍化浊解毒方可能通过降低PCNA及RegI的表达的影响,逆转胃癌前病变。

Experimental study of Xiaoguishao Decoction on PCNA and Gene Reg in rats with gastric precancerous lesions

AIM To explore the effect of Xiaoguishao Decoction(Angelicae sinensis Radix,Paeoniae Radix alba,Actinidiae argutae Radix,Trichosanthis Fructus,Poria,etc.) on gastric carcinoma precancerous lesion.METHODS One hundred normal rats were randomly divided into group A(blank control),group B(model group),group C(Xiaoguishao Decoction with large dosage group),group D(Xiaoguishao Decoction with small dosage group),group E(positive control group) with 20 rats in each group.By using N-methyl-N-nitro-N-nitrosoguanidine in combination with irregular diet,all rats but ones of group A were induced to produce gastric precancerous lesions.After creation of models,group A were feeding normally.The other groups were intragastrically administrated with distilled water,suspension of Xiaoguishao Decoction with large dosage 30 g herb’s extract,suspension of Xiaoguishao Decoction with small dosage 20 g herb’s extract,suspension of Weifuchun Tablets,respectively.Then the levels of proliferating cell nuclear antigen(PCNA) and RegI in gastric tissues were tested.RESULTS(1)The levels of PCNA of group C and D were significantly reduced,and there was significant difference(P<0.05) compared with those of group E.(2)The levels of RegI of group C were significantly reduced,and there was significant difference(P<0.05) compared with those of group E.CONCLUSION The experiment shows that reversal effect of Xiaoguishao Decoction on gastric precancerous lesions is produced.
