地肤子抗真菌化学成分研究

摘    要:目的研究地肤子抗真菌活性部位化学成分及其体外抗白色念珠菌的活性。方法采用超临界CO2萃取与不同溶剂依次提取的方法,制备地肤子的不同部位,体外实验显示其正丁醇提取部位对白色念珠菌具有明显的抑菌活性,由此采用多种色谱方法对其正丁醇提取部位进行化学成分的分离纯化,并通过波谱数据及理化性质分析对分离得到的化合物进行结构鉴定。结果从地肤子正丁醇部位分离得到8个化合物,分别鉴定为β-谷甾醇(1),β-胡萝卜苷(2),槲皮素(3),金丝桃苷(4),3-O-[β-D-吡喃木糖(1→3)β-D-吡喃葡萄糖醛酸]齐墩果酸(也称地肤子皂苷ⅠC)(5),异鼠李素-3-O-β-D-吡喃葡萄糖苷(6),3-O-[β-D-吡喃木糖(1→3)β-D-吡喃葡萄糖醛酸]齐墩果酸-28-O-β-D-吡喃葡萄糖苷(也称地肤子皂苷ⅡC)(7),3-O-[β-D-吡喃葡萄糖(1→2)β-D-吡喃木糖(1→3)β-D-吡喃葡萄糖醛酸]齐墩果酸(8)。结论化合物3、4、6均为首次从该植物中分离得到。体外抗真菌活性测试显示,化合物8对白色念珠菌有一定抑制作用。

Abstract:AIM To study the chemical constituents of the antifungal extract from Kochiae Fructus and their antifungal activity against Candida albicans in vitro.METHODS The different fractions were extracted from Kochiae Fructus by SPE-CO2 and different solvents in order.The n-butanol fraction was indicated to be antifungal against Candida albicans in vitro.The compounds were isolated by different chromatographic methods and their structures were elucidated mainly by spectral data analysis as well as physico-chemical properties.RESULTS Eight compounds were identified as β-sitosterol(1),daucosterol(2),quercetin(3),hyperoside(4),momordin Ic(5),isorhamnetin-3-O-β-D-galactoside(6),momordin IIc(7) and 2′-O-β-D-glucopyranosyl momordin Ic(8),respectively.CONCLUSION Compound 3,4 and 6 were isolated from this plant for the first time.Compound 8 could inhibit the growth of Candida albicans in vitro.
