响应曲面法优化防己中汉防己甲素及汉防己乙素的提取工艺

摘    要:目的:以汉防己甲素和汉防己乙素的提取率作为考察指标,优化防己中汉防己甲素和汉防己乙素的提取工艺。方法:利用高效液相色谱法测定汉防己甲素和汉防己乙素的含量,采用响应曲面法考察乙醇体积分数、料液比及超声时间对提取效果的影响。结果:Design expert 10预测得到汉防己甲素最佳提取工艺为乙醇浓度为60%,料液比为1∶36.07,超声时间为57.12 min,提取率为13.53 mg/g;最佳提取汉防己乙素的提取工艺为乙醇浓度62.49%,料液比为1∶37.71,超声时间为59.38 min,提取率为8.71 mg/g。结论:该研究为防己中防己甲素和汉防己乙素的提取工艺提供实验依据。

Abstract:Objective To optimize operation conditions for tetrandrine and hanfangichin B from Stephaniatetrandra S.Moore using yields as index.Methods HPLC was used to determine the concentrations of tetrandrine and hanfangichin B. Operation condition(ethanol volume fraction, solid-liquids ratio and ultrasonic time) were optimized with response surface methodology. Results According to the Design Expert 10 software as follows:the optimum operating paramaters were: ethanol volume fraction were 60% and 62.49%, solid-liquids ratio were 1∶36.07 and 1∶37.71, ultrasonic time were 57.12 min and 59.38 min for tetrandrine and hanfangichin B. The highest extraction rate of tetrandrine and hanfangichin B were 13.53 mg/g and 8.71 mg/g. Conclusion The optimized conditions are reliable and provide experiment evidences for the optimal extraction technology for tetrandrine and hanfangichin B from Stephaniatetrandra S.Moore.
