陈皮总黄酮的提取纯化工艺研究

摘    要:目的 探讨陈皮总黄酮提取、纯化工艺参数的优化方式.方法 采用紫外分光光度法以总黄酮为指标,通过正交设计试验优化陈皮总黄酮的提取工艺,同时采用大孔树脂技术优化富集总黄酮的参数指标.结果 筛选出大孔树脂分离纯化陈皮总黄酮的最佳工艺为:AB-8型大孔树脂,洗脱溶剂为50%乙醇,比上柱量为0.5g/ml,上样液浓度为0.4g生药/ml,树脂柱径高比为1:5,洗脱流速为2 BV/h,洗脱体积为5 BV.结论 纯化后总黄酮的含量明显提高.

Abstract:Objective To set up a method for extracting and purifying of flavonoids in Dried Tangerine peel.Methods Total flavonids being used as an index, UV spectrophotometer was used to optimize the extracting technology with the help of orthogonal experiment. Meanwhile macroporous resin was adopted for optimizing indexes of total flavonoids.Results The optimum conditions for purification were as followings: AB-8 macroporous resin, 50% of ethanol serving as eluting solvent, 0.4g/mL of sample concentration, 1:5 ration of diameter to height, 2BV/h of current velocity, and 5BV of flow column. Conclusion The content of total flavonoids was remarkably increased after purification.
