健康成年志愿者盐酸洛非西定的耐受性试验

本文通过对２０例健康成年志愿者服用不同剂量盐酸洛非西定后不良反应尤其是对血压的连续观察，发现服用单剂量盐酸洛非西定后１ｈ开始出现口干、乏力、困倦等不良反应，４－５ｈ达到高峰，７－８ｈ后基本消失。盐酸洛非西定可降低正常成年人卧位和立位血压，降低立位血压的作用更为突出。此外，盐酸洛非西定也有降低卧位心率的作用。盐酸洛非西定的不良反应有随剂量加大而加重的倾向。

A RESEARCH ON TOLERANCE OF HEALTHY ADULT VOLUNTEER TO LOFEXIDINE HYDROCHLORIDE

The tolerance of healthy adults to lofexidine hydrochloride(LH), an anti-opioid withdrawal drug, was studied. 20 healthy adult volunteers were randomly divided into 5 groups, one group took multi-doses-0.8 mg, tid, for two days, the other four groups took 0.2 mg, 0.4 mg, 0.6 mg and 0.8 mg, single dose separately. The side effects, mainly tiredness, drowsy, appeared one hour later after administration of LH and became most serious 4-5 hours after administration. The side effects got severer and lasted longer when the dose is increased.
