摘 要: | 目的探讨游离脂肪酸(Free Fatty Acids,FFAs)对人近曲肾小管上皮细胞(HK-2)的影响及作用机制。方法用含有不同浓度(0、125、250、500μmol/L)软脂酸的DMEM—F12培养人近曲肾小管上皮细胞(HK-2)在0h、24h、48h三个时段:MTT法检测不同环境下FFAs对HK-2细胞增殖的影响,流式细胞术检测FFAs对细胞凋亡的影响,免疫细胞化学法检测处理前后HK-2细胞中诱导型一氧化氮合酶(iNOS)及凋亡相关蛋白caspase8蛋白表达的变化。结果FFAs呈剂量和时间依赖性抑制HK-2细胞的增殖(P〈0.05),并可诱导细胞凋亡(P〈0.05)。经FFAs作用后,iNOS及Caspase8的表达增加。结论游离脂肪酸有抑制HK-2细胞增殖,促进其凋亡的作用,FFAs可能通过激活肾小管上皮细胞内的诱导型一氧化氮合酶(iNOS),加剧肾小管上皮细胞的凋亡,而caspase8参与了其发生和发展的过程。

Abstract: | Objective To explore the influence and function mechanism of free fatty acid (FFAs) to proximal convoluted tubules ep- ithelial cells(HK-2). Methods People proximal convoluted tubules epithelial ceils(HK-2),were cultured with DMEM-F12 of different concentrations of palmitic acid (0,125,250,500 μmol/L)(using defat bovine serum albumin as vector), normal medium as control group on the 0h.24h.48h respectively:applyied four methyl azo thiazole blue(determined by MTT) colorimetric detection different environment HK-2 cell proliferation capacity;Flow cytometric intraoperativewas used to observe apoptosis influence.Immune cell chemical method before and after treatment were detected HK-2 ceils iNOS and caspase8 protein expression changes. Results A FFAs doses and time dependence inhibited promoting HK-2 cell proliferation (P〈0.05),its apoptosis rate rised,compared with control signiilcantly increased (P〈0.05).When the FFAs was working, and apoptosis related proteins iNOS Caspase8 expression increased. Conclusion Free fatty acid had inhibitory HK-2 cell proliferation and promoted its apoptosis rule,FFAs may through activating renal tubular epithelial cells within the iNOS,increased renal tubular epithelial cell apoptosis,but caspase8 participated in its occurrence and development process. |
