基于NLRP3炎性小体通路探讨金樱子乙醇提取物对系膜增生性急性肾小球肾炎大鼠的保护作用

摘    要:目的:基于NLRP3炎性小体通路探讨金樱子乙醇提取物对系膜增生性急性肾小球肾炎大鼠的保护作用。方法:SD大鼠80只分为对照组、模型组、贝那普利组、金樱子组,系膜增生性急性肾小球肾炎模型建模成功后,贝那普利组、金樱子组给予相应药物灌胃,对照组、模型组给予生理盐水,持续7周。试验结束后处死大鼠,测定肾/体比值、24 h尿蛋白、血清肌酐(SCr)、血尿素氨(BUN)、肾小球系膜细胞(GMC)计数、细胞外基质(ECM)阳性面积比值、核因子κB(NF-κB)、Nod样受体家族组成的多蛋白复合物样受体家族3(NLRP3)mRNA以及蛋白水平、白细胞介素-2(IL-2)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)蛋白水平。结果:与对照组比较,模型组肾/体比值、24 h尿蛋白、SCr、BUN、GMC计数、ECM阳性面积比值、NF-κB、NLRP3 mRNA、蛋白、IL-2、IL-6、TNF-α水平明显升高(P<0.05);与模型组比较,贝那普利组、金樱子组肾/体比值、24 h尿蛋白、SCr、BUN、GMC计数、ECM阳性面积比值、NF-κB、NLRP3 mRNA、蛋白、IL-2、IL-6、TNF-α水平明显降低(P<0.05);金樱子组各指标与贝那普利组比较,差异无统计学意义(P>0.05)。结论:金樱子对大鼠系膜增生性急性肾小球肾炎具有保护作用,能明显降低肾小球系膜细胞增殖和细胞外基质沉积;其机制与金樱子能抑制NLRP3炎性小体通路NF-κB、NLRP3 mRNA、蛋白表达水平进而抑制炎症因子IL-2、IL-6、TNF-α的表达有关。

Abstract:Objective:To investigate the protective effect of ethanol extract of Rosa laevigata on acute mesangial proliferative glomerulonephritis in rats based on NLRP3 inflammatory corpuscle pathway.Methods:Eighty SD rats were divided into the control group,the model group,benazepril group,and Rosa laevigata group.Rats were made into models and received the corresponding treatment.Seven weeks later,the kidney/body weight ratio,24-hour urinary protein,SCr,BUN and GMC count,ECM positive area ratio,mRNA and protein levels of NF-κB and NLRP3,as well as the protein levels of IL-2,IL-6 and TNF-αwere determined.Results:Compared with the control group,the kidney/body weight ratio,24-hour urinary protein,SCr,BUN,GMC count,ECM positive area ratio,mRNA and protein levels of NF-κB and NLRP3,as well as the protein levels of IL-2,IL-6 and TNF-αin model group significantly increased(P<0.05).Compared with the model group,the kidney/body weight ratio,24-hour urinary protein,SCr,BUN,GMC count,ECM positive area ratio,mRNA and protein levels of NF-κB and NLRP3,as well as the protein levels of IL-2,IL-6 and TNF-αin benazepril group and Rosa laevigata group significantly decreased(P<0.05).Meanwhile the difference between the benazepril group and Rosa laevigata group was not statistic(P>0.05).Conclusion:The protective effect of ethanol extract of Rosa laevigata on acute mesangial proliferative glomerulonephritis in rats can significantly reduce the proliferation of mesangial cells and the deposition of extracellular matrix.The mechanism is related to the inhibition of the expression levels of NLRP3 inflammatory corpuscle pathway related factors including NF-κB and NLRP3 as well as the inhibition of the expressions of inflammatory cytokines including IL-2,IL-6,and TNF-α.
