Cyclin D1、Rb和P16基因在脑星形细胞瘤中表达的研究

摘    要: 目的通过检测脑星形细胞瘤组织中Cyclin D1、Rb和P16基因蛋白表达,分析其表达变化与不同病理类型肿瘤的相关性.方法选用54例Ⅰ~Ⅳ级星形细胞瘤手术后存档蜡块标本为实验标本,应用流式细胞术(FCM)分别定量检测Ⅰ~Ⅳ级星形细胞瘤组织中Cyclin D1、Rb和P16基因蛋白表达量.结果Cyclin D1蛋白的表达量随肿瘤恶性度的升高而显著增加,与病理组织学分级呈明显相关性(P<0.01);Rb蛋白和P16蛋白表达量(F1值)在低恶性组升高,高恶性组降低,两组相比差异有显著性意义(P<0.05或P<0.01).结论Cyclin D1基因蛋白表达随星形细胞瘤恶性度升高而增强;Rb和P16在高恶性度星形细胞瘤均有不同程度的表达缺失,说明细胞周期调节因子的改变在肿瘤的演化中起重要作用.

Abstract:Objective To investigate the expressions of cyclin D1,Rb and P16 gene proteins in astrocytoma,and the correlation between the changes in protein expressions and gliomas of various histopathological grades.Methods The experimental material consisted of 54 paraffin-embeded samples of brain tumor taken from patients with astrocytoma of I-IV grades.The expressions of cyclin D1,Rb and P16 gene proteins in astrocytoma were detected quantitatively by flow Cytometry (FCM).Results The expression of cyclin D1 protein in astrocytoma increased and correlated with increasing histological grade (P<0 01).The expression of both Rb and P16 proteins were higher in lower grade group,but lower in high grade group,with significant difference between the two groups (P<0 01,P<0 05) Conclusions The expression of cyclin D1 positively correlates with the degree of malignancy,and the negative expressions of Rb protein and P16 protein were detected mainly in high degree astrocytoma,suggesting that gene alterations of cell cycle regulator may play an important role in the development of astrocytoma.
