miR-1246在肝癌组织中的表达及其对人肝癌细胞增殖和凋亡的影响

摘    要:
目的 观察微小RNA-1246(miR-1246)在肝癌组织中的表达及其对人肝癌细胞增殖和凋亡的影响,探讨其在肝癌发生发展中的作用和意义。方法应用实时定量PCR(qRT-PCR)方法检测miR-1246在肝癌以及癌旁组织的表达;对人肝癌细胞株(BEL7402、SMMC7721)转染miR-1246 抑制剂后,采用MTT法观察肝癌细胞的增殖活性,流式细胞仪检测细胞凋亡的变化。结果 miR-1246 在肝癌组织中表达显著高于癌旁组织[(65.39±8.77)vs(10.23±2.58),P=0.002]。MTT和凋亡实验结果显示,与阴性对照组相比,转染miR-1246抑制剂后,BEL7402细胞增殖能力下降,凋亡增加(P均<0.05);SMMC7721细胞也出现增殖能力下降,凋亡增加(P均<0.05)。结论 miR-1246 在肝癌组织中高表达,并且与肝癌细胞的增殖及凋亡有关,其表达的上调可能与肝癌的发生发展有关。

Abstract:
objective To observe the expression of miR-1246 in human hepatocellular carcinoma tissues and its effect on cell proliferation and apoptosis. Methods The expression of miR-1246 in hepatocellular carcinoma tissues and normal liver tissues was detected by real-time PCR (qRT-PCR). The cultured hepatocellular carci-noma cell line BEL7402 and SMMC7721 cells were transfected with miR-1246 inhibitor, the cell proliferation was detected by MTT assay and apoptosis was analyzed by flow cytometry. Results The expression of miR-1246 in hepatocellular carcinoma tissues was significantly higher than that in normal liver tissues [(65.39±8.77) vs (10.23±2.58), P=0.002)]. After transfected miR-1246 inhibitor, BEL7402 cell transfected hepatocellular carci-noma cells showed low proliferation rate and high apoptosis rate when compared with control group (P<0.05), the same trend was also observed in SMMC7721 cells (P<0.05). Conclusion These results indicate that the miR-1246 is highly expressed in hepatocellular carcinoma tissues and its high expression may be involved in the de-velopment of hepatocellular carcinoma.
