脑梗死大鼠外周血中CD4^+CD25^+调节性T细胞表达对神经功能变化的影响

摘    要:目的观察脑梗死大鼠建模成功后不同时期CD4^+CD25^+调节性T细胞(CD4^+CD25^+Treg)表达对神经功能变化情况及两者之间的相关性。方法选择清洁级雄性大鼠60只,适应性饲养1 w后,选择30只作为假手术组,另外30只建立大脑中动脉闭塞(MCAO)模型,分别于建模成功后12、24、72 h后处死大鼠,每次处死10只。检测并对比建模成功后不同时期大鼠CD4^+CD25^+Treg表达与神经功能(以Zausinger六分法评估大鼠神经功能),并分析上述两者之间的相关性。结果建模成功12 h后,模型组神经功能得分显著低于假手术组(P<0.05);两组CD4^+CD25^+Treg比较差异无统计学意义(P>0.05);建模成功24、72 h后,模型组CD4^+CD25^+Treg与神经功能得分显著低于假手术组(P<0.001);经双变量Pearson直线相关性检验结果显示,脑梗死大鼠CD4^+CD25^+Treg与神经功能呈正相关(r=0.593,P=0.001)。结论脑梗死大鼠梗死早期CD4^+CD25^+Treg未见明显改变,随着梗死时间延长,大鼠CD4^+CD25^+Treg逐渐降低、神经功能损伤程度加重,二者间存在相关性,考虑早期通过检测脑梗死CD4^+CD25^+Treg,对预测神经功能、指导治疗有积极意义。

Effects of expression of CD4^+CD25^+regulatory T cells in peripheral blood on changes of nerve function in rats with cerebral infarction

Abstract:Objective To observe the expression of CD4^+CD25^+regulatory T cells(CD4^+CD25^+Treg)on neural function changes in different periods after successful modeling of cerebral infarction rats and analyze the correlation between CD4^+CD25^+Treg and neural function changes.Methods 60 clean male rats were selected.After one week of adaptive feeding,30 rats were selected as the sham group.Another 30 rats were established into the MCAO model,sacrificed after 12 h,24 h,and 72 h of successful modeling and 10 rats were sacrificed each time.Expression of CD4^+CD25^+Treg and neural function were detected and compared at different stages after successful modeling and the correlation between expression of CD4^+CD25^+Treg and neural function was analyzed.Results After 12 hours of successful modeling,the neurological function score of model group was significantly lower than that of sham group(P<0.05).There was no significant difference in expression of CD4^+CD25^+Treg between model group and sham group(P>0.05).After the successful modeling of 24 hours and 72 hours,CD4^+CD25^+Treg and neurological function score of model group were lower than those of sham group(P<0.05).Bivariate Pearson linear correlation test showed that CD4^+CD25^+Treg in cerebral infarction rats was positively correlated with neural function(r=0.593,P=0.001).Conclusions There is no significant change in CD4^+CD25^+Treg at the early stage of cerebral infarction in rats.With the extension of the infarction time,the CD4^+CD25^+Treg in rats gradually decrease and the degree of neurological damage increase.There is a correlation between the expression of CD4^+CD25^+Treg and neural function.Early detection of CD4^+CD25^+Treg of cerebral infarction has positive significance for predicting nerve function and guiding treatment.
