巴西天然蜂胶对牙龈卟啉单胞菌激活的巨噬细胞极化的调控及机制探讨

摘    要:
目的:探讨巴西天然蜂胶对牙龈卟啉单胞菌脂多糖(P.gLPS)激活的人巨噬细胞(THP-1)极化的调控及分子机制。方法:染色分析人慢性牙周炎牙龈组织中巨噬细胞的浸润;检测P.gLPS激活的THP-1中一氧化氮合成酶(iNOS)和精氨酸合成酶(Arginase-1)mRNA的表达;检测巴西天然蜂胶对P.gLPS激活的THP-1极化的调控及对NF-gB活化的抑制作用。结果:人慢性牙周炎牙龈组织中巨噬细胞大量浸润:P.gLPS诱导THP-1高表达iNOS,微量表达Arginase-1。巴西天然蜂胶显著抑制P.gLPS诱导THP-1表达的iNOS和TNF-α,并抑制P.gLPS诱导的NF-KB活化。结论:证实了巴西天然蜂胶通过抑制P.gLPS激活的THP-1中NF-xB的活化,阻止了THP-1向M1极化。

Brazilian Green Propolis Suppresses the Porphyromonas Gingivalis LPS-induced M1 Responses by Inhibiting NF-KBActivation in Human Macrophages.

Abstract:
Objective: To investigate the roles of propolis to P. g LPS-induced human macrophage phenotype. Methods: HE staining and DAB staining were used to evaluate the macrophage infiltration in the gingival tissues of periodontitis patients. RT-PCR was used to measure mRNA of iNOS and Arginase-1 in P: g LPS induced THP -1. RT-PCR and Elisa were used to measure the roles of propolis to P. g LPS-induced THP-1 phenotype. Western blotting and Elisa were used to evaluate the inhibitory function of propoiis on NF kB activation. ResultS: Many infiltrated macrophages were detected in the gingival tissues of periodontitis. P. g LPS(l/ug/ml) significantly increased mRNA of iNOS and little increased mRNA of Arginase-1 in THP-1. Propolis(15ug/ml) significantly, inhibited P. g LPS-increased mRNA of iNOS and secretion of TNF-a in THP-1. Propolis inhibited P.g, LPS induced NF-kB activation in THP-1. Conclusion: Propolis suppresses P. g LPS-induced M1 phenotype through inhibition on NF-kB activation.
