激素性股骨头坏死股骨头内PPARγ及BMP2的动态表达

摘    要:目的 研究激素性股骨头坏死股骨头内局部过氧化物酶体增殖物激活受体γ(peroxisome proliferators-activated receptor γ,PPARγ)和BMP2的基因表达及蛋白合成的动态变化.方法 健康成年新西兰大白兔,共35只,雌雄各半,质量2.6~3.2 kg,平均2.9 kg.完全随机的方法将实验动物分为正常对照组15只,模型组20只.用改进的马血清加甲基强的松龙的方法诱导制成兔激素性股骨头坏死动物模型,HE染色确定模型的成功建立,提取兔股骨头内总RNA及总蛋白进行实时定量PCR及Western blot检测,研究不同时段坏死股骨头内PPARγ及BMP2的基因表达、蛋白合成的动态变化特点.结果 造模后2、4、8周时,模型组动物股骨头内PPARγ的基因表达、蛋白合成较对照组上调,其中PPARγ mRNA的表达量分别为正常组的1.2583、1.4329、2.0135倍,且随时间的推移PPARγ的基因表达、蛋白合成呈进行性上调趋势.模型组动物股骨头内BMP2的基因表达、蛋白合成较对照组下降,其中BMP2 mRNA的表达量在造模后2、4、8周分别为正常组的0.3264、0.1317、0.1253倍.BMP2蛋白合成量与病程的时间呈负相关.结论 素性股骨头坏死早期,股骨头内PPARγ过度表达,而BMP2的表达受到抑制.

Genes expressions of PPARγ and BMP2 inside the femoral head of steroid-induced osteonecrosis in rabbits
Abstract:Objective To study the dynamic,changes of gene expression and protein synthesis of PPARγ(peroxisome proliferators-activated receptorγ) and BMP2 inside the femoral head of steroid-induced osteonecrosis in rabbits.Methods 35 rabbits involved in this study,including 17 females and 18 males,with the mean weight of 2.9 kg,ranged from 2.6 to 3.2 kg.All rabbits were divided into model group(20 rabbits) and control group (15 rabbits) randomly.Rabbit model of femoral head necrosis was made with the method of horse sernm plus methylprednisolone.Hematoxylin and eosin stain,extract total RNA and total protein from the femoral head for real-time quantitative PCR,and Western blot were performed to detect the dynamic of the PPAR γ and BMP2 genes expressions and protein synthesis inside the femoral head of steroid-induced osteonecrosis in rabbits.Results At the post-modeling 2th,4th and 8th week,the expression of the PPARγ gene,protein synthesis in group B(model group)were obviously higher than those in control group,and the gene expressions were 1.2583,1.4329,2.0135 times of those in control group.Its expression had an upgrade tendency,with the longer time.The expression of the BMP2 gene,protein synthesis in group B (model group) at the post-modeling 2th,4th and 8th week were extremely lower than those in control group,which were 0.3264,0.1317,0.1253 times of those in control group.And it had a negative relation with the time of disease.Conclusion Inside the femoral head of steroid-induced osteonecrosis in rabbits,the gene and protein expression of the PPARγ are upgraded;the expression of the BMP2 gene and protein are inhibited.
