白介素-8对SKOV3细胞系卵巢癌干样细胞特性的影响

摘    要:
目的:研究白介素-8(IL-8)能否促成人卵巢癌SKOV3细胞系细胞肿瘤干细胞样细胞特性.方法:用添加或未添加重组人IL-8(rhIL-8:10.0 ng/mL)的干细胞条件培养基处理SKOV3细胞.肿瘤球形成和琼脂集落形成试验测定球形成率和集落形成率.蛋白质印迹分析肿瘤干细胞标志蛋白CD133和CD44表达水平.结果:rhIL-8(10.0 ng/mL)显著地增高SKOV3细胞肿瘤球形成率和琼脂集落形成率.此外,rhIL-8(10.0 ng/mL)上调SKOV3细胞CD133和CD44蛋白表达.结论:rhIL-8具有促进SKOV3细胞肿瘤干细胞样细胞特性作用.

Abstract:
Objective To investigate whether interleukin-8(IL-8) could promote the characteristics of cancer stem-like cells from ovarian cancer SKOV3 cell line. Methods SKOV3 cells were treated with and without stem cell condition medium addition with recombination human IL-8(rhIL-8: 10.0 ng/mL). Sphere forming ratio and clone forming ratio was detected using sphere forming assay and clone forming assay. The expression of cancer stem cell marker proteins including CD133 and CD44 was analyzed using western blot. Results rhIL-8(10.0 ng/mL) significantly increased sphere forming ratio and clone forming ratio in SKOV3 cells. Moreover, the expression of CD133 and CD44 was up-regulated by rhIL-8(10.0 ng/mL). Conclusion rhIL-8 could promote the cancer stem like cells characteristics in SKOV3 cells.
